摘  要:目的研究乙醛脱氢酶1A1(ALDH1A1)蛋白的结构与功能,进一步探究其在动脉粥状硬化发生发展及临床病理因素的关系。方法利用Protparam、ProtScale、NetPhos3.1、YinOYang 1.2 Server、NetNGlyc 1.0Server、SOPMA及SWISS-MODEL等在线软件和生物信息学手段对ALDH1A1蛋白的理化性质、亲疏水性、磷酸化位点、糖基化位点及二、三级结构等生物学特性进行了系统分析,并利用Clustal×2和MEGA7.0软件对ALDH1A1蛋白进行同源性分析和系统进化分析。结果ALDH1A1蛋白由563个氨基酸组成,是亲水碱性蛋白,不稳定指数为41.65;在体外哺乳动物中的半衰期为30h;共有44个可能的磷酸化位点和8个可能的糖基化位点;无跨膜螺旋区与信号肽;二级结构预测结果显示,ALDH1A1蛋白中α-螺旋结构与无规则卷曲结构占比都较高,而β-转角结构占比很少,只有4.97%;同源性与进化性分析显示,与褐家鼠序列一致性最高的是黄金仓鼠,它们的亲缘关系最近。结论ALDH4A1进化性高度保守功能重要,有助于揭示蛋白功能、靶向药物设计,有助于动脉粥样硬化治疗提供理论参考。Objective To study the structure and function of acetaldehyde dehydrogenase 1A1(ALDH1A1)protein,and to further explore its relationship with the development and clinicopathological factors of arteriosclerosis.Methods Protparam,ProtScale,NetPhos3.1,YinOYang1.2 Server and NetNGlyc 1.0 were used Server,SOPMA,Swiss-Model and other online software and bioinformatics methods were used to systematically analyze the physical and chemical properties,hydrophobicity,phosphorylation sites,glycosylation sites,secondary and tertiary structure of ALDH1A1 protein.The homology and phylogenetic analysis of ALDH1A1 protein were performed by Clustal×2 and MEGA7.0 software.Results ALDH1A1 protein was composed of 563 amino acids and was hydrophilic and basic.The instability index was 41.65.In vitro half-life of mammals is 30 h;There were44 possible phosphorylation sites and 8 possible glycosylation sites.No transmembrane helical region and signal peptide;The secondary structure prediction results showed that the proportion ofα-helix structure and irregular curl structure in ALDH1A1 protein was high,while the proportion ofβ-turn structure was low(only 4.97%).Homology and evolutionary analysis showed that the highest sequence consistency with Rattus norvegicus was the golden hamster,which was the most closely related.Conclusion The evolutionary-highly conserved function of ALDH4 A1 is important,which can help to reveal the protein function,target drug design,and provide theoretical reference for the treatment of atherosclerosis.
