摘  要:为了探讨ISSR和SSR标记在杂交野大麦新品系鉴定中的可行性,用12个ISSR引物和6对SSR引物对两个新品系进行指纹图谱的鉴定。结果表明,ISSR标记引物和SSR标记引物的平均多态性位点百分率分别为77.93%和69.22%,均表现出较高的多态性;两种标记的遗传相似系数和聚类分析表明,杂交新品系Y_(33)和P_(13)偏向轮回亲本野大麦遗传;两种标记不同引物的不同扩增片段相互组合都可将供试材料区别开。ISSR和SSR标记的指纹图谱都能客观真实地检测出亲本与杂交后代间的遗传差异,可用于野大麦远缘杂种鉴定,并为新品种审定提供依据。In order to study the feasibility of ISSR and SSR markers in the identification of new lines of hybrid Hordeum brevisubulatum,12 ISSR primers and 6 pairs of SSR primer were used to identify the fingerprints of the two new lines.The results showed that the average percentage of polymorphic loci of ISSR labeled primers and SSR labeled primers were 77.93%and 69.22%,respectively,both of which exhibited high polymorphism;The genetic similarity coefficient and cluster analysis of the two markers showed that the new hybrid lines Y_(33) and P_(13) were biased to the recurrent parent Hordeum brevisubulatum;The combination of two different fragments labeled with different primers could distinguish the materials.The fingerprints of ISSR and SSR markers could objectively and truly detect the genetic differences between parents and hybrid offspring,which could be used for the identification of distant hybrids of Hordeum brevisubulatum and provide basis for the approval of new varieties.
