摘  要:目的预测和分析沙眼衣原体主要外膜蛋白(MOMP)的免疫优势表位。方法以UniProt蛋白质数据库中检索CtMOMP的氨基酸序列为基础,针对MOMP的理化性质、二级结构、亲水性参数、表面可及性参数、极性参数、抗原性和柔韧性参数,预测其B细胞表位;用SYFPEITHI、IEDB及NetCTI 1.2 server预测MOMP的细胞毒性T淋巴细胞(CTL)表位;用RANKPEP及SYFPEITHI预测MOMP的辅助T细胞(Th)表位,筛选分值共同较高的多肽片段作为候选表位。应用GalaxyWEB对MOMP进行三维结构建模分析。结果MOMP由393个氨基酸残基组成,以无规则卷曲及α螺旋为主;推测MOMP的B细胞表位位于N端42~52 aa、161~179 aa、259~265 aa和349~358 aa区段;其CTL表位主要集中于近C端,且以HLA-A*02:01、HLA-A*03限制性表位为主;Th表位各表型候选表位肽数目均较多。结论沙眼衣原体MOMP含有潜在的B细胞和T细胞抗原表位,其免疫优势表位主要位于N端42~65 aa、155~185 aa区段和C端342~359 aa区段。Objective To predict and analyze the immunodominant epitopes of major outer membrane protein(MOMP)of Chlamydia trachomatis(Ct).Methods The amino acid sequence of Ct MOMP was retrieved from UniProt protein database,and the B-cell epitopes of MOMP were predicted according to its physical and chemical properties,sec-ondary structure,hydrophilicity parameters,surface accessibility parameters,polarity parameters,antigenicity and flexi-bility parameters.Cytotoxic T lymphocyte epitopes were predicted by SYFPEITHI,IEDB,and Net CTI 1.2 server.Help-er T cell epitopes were predicted by RANKPEP and SYFPEITHI,and the polypeptide fragments with high scores were se-lected as the candidate epitopes.Meanwhile,the three-dimensional structure of MOMP was modeled by Galaxy web.Re⁃sults MOMP was composed of 393 amino acids,mainly random coil andαhelix;it was speculated that B-cell epitopes were located in the N-terminal 42-52aa,161-179aa,259-265aa and 349-358aa segments;its CTL epitopes were mainly concentrated in the proximal C-terminal,and mainly HLA-A*02:01 and HLA-A*03 restricted epitopes.The number of candidate epitope peptides predicted by each phenotype of Th epitope was higher.Conclusion Bioinformatics analysis shows that MOMP contains potential B-cell and T-cell epitopes,and its immunodominant epitopes are mainly located in the N-terminal 42-65aa,155-185aa and C-terminal 342-359aa.
