摘 要:为了解决化学药品滥用而带来的病虫害抗性问题,笔者以6种不同化学型的墨西哥柏木叶精油为试验对象,采用清除2,2′-叠氮基双(3-乙基苯并噻唑-6-磺酸)二铵盐(ABTS^(+))和2,2-二苯基-1-吡啶并肼基(DPPH)自由基的方法,探讨其体外抗氧化能力;通过抑菌圈大小和最小抑菌杀菌浓度来判断墨西哥柏木叶精油的杀菌抑菌活性;通过对秀丽隐杆线虫的毒性来评估其杀虫驱虫活性。结果表明,香紫苏型对ABTS^(+)自由基清除效果最佳,其清除率达10%时所用精油的质量浓度(LC_(10))为1.0134 mg/mL,依兰油型对DPPH自由基清除效果最佳(LC_(10)为0.8931 mg/mL)。香紫苏型和依兰油型对枯草芽孢杆菌的抑制效果最好,当体积分数为100%时,抑菌圈直径约为11 mm,最小抑菌浓度和最小杀菌浓度分别为3.75和7.50 mg/mL。高浓度时香紫苏型对线虫毒杀效果最佳,且能很好地破坏线虫体内的氧化平衡,加速线虫死亡;100 mg/L质量浓度下,柠檬烯型可抑制过氧化氢酶活性,香紫苏型可抑制谷胱甘肽酶活性,蒎烯型可抑制超氧化物歧化酶活性。研究结果表明,墨西哥柏木叶精油对秀丽隐杆线虫体内抗氧化酶活性有很大影响,能缩短秀丽隐杆线虫的寿命。在开发环境友好型杀菌剂和杀虫剂时,可以使用墨西哥柏木叶精油作为增效剂,以提高其综合杀灭能力。Cupressus lusitanice is a species of cypress, which is fast-growing and drought-resistant, and also demonstrates resistance properties against insects. Due to these properties, it is widely used as timber around the world. The stems and leaves of C. lusitanice are rich in essential oils, and the oils have antibacterial, bactericidal, insecticidal and repellent effects. In order to solve the problems of pest resistance caused by the chemical abuse, this study used six essential oils from C. lusitanice leaves to investigate their capacities of antioxidant, antibacterial and toxicity to Caenorhabditis elegans. The antioxidant activity in vitro was investigated by scavenging ABTS^(+)and DPPH free radicals. The bactericidal activity of the essential oils from C. lusitanice leaves were judged by the inhibition zone, minimum inhibitory concentration and minimum bactericidal concentration. The insecticidal activity of essential oils from C. lusitanice leaves were evaluated by their toxicity to C. elegans. The Sclareol type oil was the most efficient for removing ABTS;free radicals, when the scavenging rate reached 10%, the mass concentration of the essential oil was 1.013 4 mg/mL, while the Murolene type oil was the most efficient for removing DPPH free radicals, when the scavenging rate reached 10%, the mass concentration of the essential oil was 0.893 1 mg/mL. The Sclareol and Murolene oils were equally good for inhibiting Bacillus subtilis when the volume concentration was 100%, producing an 11 mm inhibition zone, with a minimum inhibitory concentration of 3.75 mg/mL, and a minimum bactericidal concentration of 7.50 mg/mL. At high concentrations, the toxic effect of the Sclareol type on C. elegans was the best;At the same time, it can destroy the oxidation balance of C. elegans and accelerate the death of C. elegans.
