摘 要:以贵州省生物研究所保藏的鲜艳乳菇菌株为试材,研究了不同氮源、碳源和无机盐对菌丝生长的影响,筛选出适宜菌丝生长的营养成分,并结合正交试验,对母种培养基配方进行优化。结果表明,酵母浸粉作为氮源加快菌丝生长速度,牛肉浸粉培养下菌丝密度增加,酵母浸粉和牛肉浸粉(质量比7∶3)的组合氮源比单一氮源更有利于菌丝生长,葡萄糖是最适宜的碳源,CaCl_(2)能显著促进菌丝生长。鲜艳乳菇母种培养基优化配方为:酵母浸粉3.5 g/L、牛肉浸粉1.5 g/L、葡萄糖20 g/L、CaCl_(2)1.5 g/L、琼脂18~20 g/L,菌丝生长速度达2.18 mm/d。The test material Lactarius vividus strains was preserved by the Guizhou Institute of Biology. The effects of different nitrogen sources, carbon sources and inorganic salts on the mycelia growth were studied, and the suitable nutrients for the mycelia growth were selected. With the orthogonal test, medium formula was optimized. The results showed that, as nitrogen source, yeast extract powder could accelerate the growth rate of mycelia. The mycelial density was increased in beef extract powder culture. The combined nitrogen source of yeast extract powder and beef extract powder(mass ratio: 7∶3) was more favorable to the growth of mycelia than the single nitrogen source. Glucose was the most suitable carbon source. CaCl_(2)could significantly promote the growth of mycelia. The best medium formula of Lactarius vividus was that 3.5 g/L yeast extract powder, 1.5 g/L beef extract powder, 20 g/L glucose, 1.5 g/L CaCl_(2)and 18-20 g/L agar. The growth rate of mycelia was 2.18 mm/d.
