摘 要:目的建立高效液相色谱(HPLC)法同时测定胡颓子叶中槲皮素、银锻苷和山柰酚的含量,为胡颓子叶中质量标准的提升奠定基础。方法采用Kromasil Eternity-5C 18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.05%磷酸水溶液(54∶46)为流动相洗脱,流速1.0 mL/min,柱温30℃,检测波长360 nm。结果槲皮素、银锻苷、山柰酚分别在0.010~0.522μg(r=0.9999)、0.021~1.051μg(r=0.9999)、0.021~1.048μg(r=0.9999)时与峰面积线性关系良好;平均加样回收率(n=3)均在98.4%~98.7%,相对标准偏差(RSD)均小于1.3%。结论HPLC法简便、准确、重复性好,可用于胡颓子叶中槲皮素、银锻苷、山柰酚的质量控制。Objective To establish a high performance liquid chromatography(HPLC)method for the simultaneous determination of quercetin,tiliroside and kaempferol in folium elaeagni,so as to lay a foundation for the improvement of quality standards in folium elaeagni.Methods The Kromasil Eternity-5-C 18(250 mm×4.6 mm,5μm)chromatographic column was used,and methanol-0.05%phosphoric acid aqueous solution(54∶46)was used as the mobile phase.The flow rate was 1.0 mL/min,the column temperature was 30℃,and the detection wave length was 360 nm.Results The quercetin,tiliroside and kaempferol had good linear correlation with peak area at 0.010-0.522μg(r=0.9999),0.021-1.051μg(r=0.9999),0.021-1.048μg(r=0.9999),respectively.The average sample recovery rates(n=3)were all in the range from 98.4%to 98.7%.And the relative standard deviations(RSD)were all less than 1.3%.Conclusion HPLC method is simple,accurate and reproducible.It can be used for the quality control of quercetin,tiliroside and kaempferol in folium elaeagni.
