摘 要:目的 探究加味香连丸对急性放射性肠炎大鼠核转录因子-κB(NF-κB)信号通路的影响。方法 将40只SPF级SD大鼠随机分为正常组、模型组、加味香连丸组、思密达组。对除正常组外的3组大鼠予直线加速器6-MV的X射线单次全腹部照射,总剂量12 Gy,剂量率200 cGy/min,建立急性放射性肠炎模型。照射后12 h开始灌胃,观察各组大鼠形态学、体质量变化;利用光学显微镜下观察及图像分析软件对比各组肠组织病理改变,并用ELISA法测定肠组织Toll样受体4(TLR-4)、NF-κB p65、白细胞介素(IL)-6、肿瘤坏死因子-α(TNF-α)含量。结果 模型大鼠均出现急性放射性肠炎症状,小肠组织不同程度地出血、坏死。加味香连丸组TLR-4、NF-κB p65及相关炎症因子IL-6、TNF-α含量与正常组比较未见明显差异(P> 0.05)。结论 加味香连丸对急性放射性肠炎大鼠NF-κB信号通路存在影响,能改善相关炎症指标,阻止炎症信号放大,对肠道黏膜起到一定的保护作用。Objective:To explore the effect of Jiawei Xianglian Pills on NF-κB signaling pathway in rats with acute radiation enteritis. Methods:40 SPF-grade SD rats were randomly divided into the normal group,control group,Jiawei Xianglian Pills group and Smecta group. The acute radiation enteritis model was established by single total abdominal irradiation of 6-MV X-ray at the linear accelerator with a total dose of 12 Gy and a dose rate of 200 cGy/min. Each group received intragastric administration 12 h after radiation. The changes of morphology and weight of rats were observed. The pathological changes of intestinal tissues were compared by optical microscope and image analysis software,and the levels of TLR-4,NF-κB p65,IL-6 and TNF-α were measured by ELISA. Results:Acute radiation enteritis model was found in groups except the normal one,presenting pathological manifestations with bleeding and necrosis of the small intestine. ELISA results showed that there were no significant differences in the levels of TLR-4,NF-κB p65 and inflammation-associated cytokines IL-6 and TNF-α between the Jiawei Xianglian Pills group and the normal group(P > 0.05). Conclusion:Jiawei Xianglian Pills can affect NF-κB signaling pathway in rats with acute radiation enteritis,improve inflammation-associated indicators,prevent amplification of inflammatory signals,and protect intestinal mucosa to some extent.
