摘 要:【目的】开展苦瓜不同种质果肉厚度统计分析以及转录组测序分析,为后续研究苦瓜果实发育及相关调控机制提供理论参考。【方法】以70份苦瓜种质为对象,测定分析果肉厚度,并对差异显著材料进行高通量转录组测序和数据分析。【结果】(1)70份苦瓜材料果肉厚度存在品系间差异,其中LX1-3和ZK54差异显著,将前者作为厚肉型后者作为薄肉型材料用于转录组分析;(2)经转录组测序统计,LX1-3共得到得到234825400条干净序列,平均Q30值为92.74%,比对到参考基因组上的序列占总序列数的88.07%,其中,单一比对序列34027357条,占比86.98%,GC含量为46.54%;ZK54共得到241318020条干净序列,平均Q30值为92.26%;比对到参考基因组上的序列占总序列数的87.23%,其中,单一比对序列34607181条,占比86.06%,GC含量为46.75%;(3)2份种质中共发现差异基因1492个,预测到598个转录因子,GO数据库比对共得到616个注释,包括生物过程、细胞组分和分子功能三大类,参与生物过程的最多。KEGG代谢通路共富集到308个差异基因,其中上调基因中富集因子最高的为“卟啉和叶绿素代谢”途径,富集差异基因数目最多的途径为“核糖体”;下调基因中富集因子最高和富集下调差异基因最多的通路均为“苯丙素生物合成”。【结论】利用高通量测序结果推测苦瓜果肉发育可能受上调基因表达调控影响较大,这将有利于从分子水平对苦瓜发育调控机理进行深入研究,为推动苦瓜分子育种提供参考。【Objective】Flesh thickness variation and transcriptome sequencing analysis of different bitter gourd were carried out to provide theoretical reference for subsequent studies on fruit development and related regulatory mechanisms bitter gourd.【Method】Flesh thickness of 70 bitter gourds was determined,then high-throughput transcriptome sequencing and data analysis were performed on the significantly different materials.【Result】(i)Differences of flesh thickness were found among 70 materials,and LX1-3 and ZK54 differed significantly which were applied as thick-flesh and thin-flesh germplasms to do the transcriptome sequencing.(ii)According to the statistics of transcriptomic sequencing,a total of 234825400 clean reads were obtained from LX1-3,with an average Q30 value of 92.74%.The sequences compared to the reference genome accounted for 88.07%of the total number of sequences,including 34027357 single sequence,accounting for 86.98%,and GC content was 46.54%.Then,a total of 241318020 clean reads were obtained from ZK54,with an average Q30 value of 92.26%.The sequences compared to the reference genome accounted for 87.23%of the total number of sequences,including 34607181 single sequences,accounting for 86.06%,and GC content was 46.75%.(iii)Totally 1492 differentially expressed genes were found and 598 transcription factors were predicted between these two germplasms.616 annotations were obtained in GO database,including biological processes,cell components and molecular function,therein most of them were involved in biological processes.308 different genes were enriched in KEGG metabolism pathway,‘Porphyrin and Chlorophyll metabolism’pathway was with the highest enrichment factor and‘Ribosome’was with the largest number among the up-regulated genes,‘Phenylpropanoid biosynthesis’pathway was with both the highest enrichment factor and the largest number among the down-regulated genes.【Conclusion】Based on the results of high-throughput sequencing,it is speculated that flesh development of bitt
