摘 要:【目的】比较绵羊卵母细胞体外受精后卵裂快慢对胚胎发育潜能及移植怀孕率的影响。【方法】使用因卵巢粘连无法作为超排供体的成年杜泊母羊,常规激素诱导卵泡超数发育,手术或屠宰回收卵丘—卵母细胞复合体(COCs),进行体外成熟(IVM),体外受精(IVF)和体外培养(IVC)。分别统计受精24、36、48 h卵裂情况及第7天的囊胚率,并按实验设计进行移植,统计怀孕率。其中,受精24 h及之前卵裂的记为卵裂快组,受精24 h之后卵裂的胚胎记为卵裂慢组。【结果】(1)卵裂快组胚胎占总胚胎数比例达77.85%,极显著高于卵裂慢组的22.15%(P<0.01);(2)卵裂快组囊胚率极显著高于卵裂慢组(68.95%vs 26.91%,P<0.01);(3)卵裂快组的早期胚胎移植单枚或双枚,怀孕率差异不显著(64.05%vs 65.62%,P>0.05);(4)卵裂慢组的早期胚胎移植两枚的怀孕率极显著高于移植单枚的结果(33.89%vs 11.67%,P<0.01),但均极显著低于卵裂快组的胚胎(P<0.01);(5)囊胚单、双胚移植怀孕率无显著差异(69.33%vs 70.46%,P>0.05);(6)受精后24 h或48 h移植,对怀孕率没有影响;(7)从形态上无法判断早期胚胎的质量。【结论】绵羊体外早期胚胎,卵裂快组单枚移植,卵裂慢组双枚移植,囊胚单枚移植,能提高胚胎利用率。【Objective】To compare the effects of cleavage rate of sheep oocytes after in vitro fertilization on embryonic development potential and pregnancy rate after transplantation.【Method】Adult Dorper ewes which could not be used as superovulation donors due to ovarian adhesion were selected and stimulated by conventional hormone to induce the superdevelopment of follicles.The cumulus and oocyte complexes(COCs)were recovered by operation or slaughter,for in vitro maturation(IVM),in vitro fertilization(IVF)and in vitro culture(IVC).The cleavage at 24,36 and 48 hours after fertilization and the blastocyst rate at 7 days were counted respectively,The early embryos or blastocysts were transferred according to the experimental design,and the pregnancy rate of each group was counted and statistically analyzed.Among them,the embryos cleaved at and before 24 hours post insemination was recorded as fast cleavage group,while the cleavage after 24 hours post insemination was recorded as slow cleavage group.【Result】(i)The proportion of embryos in the fast cleavage group to cleaved embryos in the total number was 77.85%,which was significantly higher than 22.15%in the slow cleavage group(P<0.01);(ii)The blastocyst rate in fast cleavage group was significantly higher than that in slow cleavage group(68.95%vs 26.91%,P<0.01);(iii)There was no significant difference in pregnancy rate between single or double early cleavage embryos transfer in fast cleavage group(64.05%vs 65.62%,P>0.05);(iv)The pregnancy rate of two early embryos transfer was significantly higher than that of single embryo in slow cleavage group(33.89%vs 11.67%,P<0.01),but was all significantly lower than that of fast cleavage embryos(P<0.01);(v)There was no significant difference between single blastocyst and double blastocyst(69.33%vs 70.46%,P>0.05);(vi)Transplantation at 24 hours or 48 hours post insemination had no effect on pregnancy rate;(vii)It was impossible to judge the quality of the early embryo from its morphology.【Conclusion】For sheep early e
