摘　　要：目的建立适合于同时测定人血浆中美罗培南与亚胺培南浓度的通用高效液相色谱快速检测方法。方法采用外标法定量,血浆样品采用适量的乙腈沉淀蛋白,离心后取上层清液加入二氯甲烷萃取,涡旋离心后取上清液,再次离心后进样分析。采用Agilent 1260高效液相色谱仪,色谱柱为Agilent ZORBAX SB-C_(8)(4.6 mm×250 mm,5μm),流动相为乙腈-0.1 mol·L^(-1)乙酸铵溶液,采用梯度洗脱法,流速为1 mL·min^(-1),柱温30℃,检测器采用二极管阵列检测器,检测波长为298 nm。结果美罗培南在1~200μg·mL^(-1)范围内线性良好(r=0.99990),亚胺培南在1~200μg·mL^(-1)范围内线性关系良好(r=0.99957);两种药物的低、中、高浓度(2.0,80.0,200.0μg·mL^(-1))的日内、日间精密度RSD在0.19%~1.53%范围之内;平均提取回收率在(81.67±6.30)%~(90.83±3.34)%范围之内;方法回收率在(86.20±1.30)%~(114.20±1.48)%范围之内。结论本方法专属性好,灵敏度高,操作简便,可满足临床美罗培南与亚胺培南血药浓度的监测的需求。OBJECTIVE To establish a rapid,commonly,reversed phase HPLC method for the determination of meropenem,imipenem concentrations in human plasma.METHODS Using external standard and plasma samples were extracted from human plasma with dichloromethane after precipitated protein by acetonitrile.And then detected on Agilent ZORBAX SB-C_(8)(4.6 mm×250 mm,5μm)with DAD detector at 298 nm.The mobile phase consisted of acetonitrile-0.1 mol·L^(-1) ammonium acetate,with a flow rate of 1 mL·min^(-1) gradient elution.RESULTS The method was validated and he meropenem concentration range from 1.00 to 200.00μg·mL^(-1)(r=0.99990);the panipenem concentration range from 1.00 to 200.00μg·mL^(-1)(r=0.99957).The method was ciliated for the low,middle and high concentrations of check samples were:intra-run and inter-run RSDs were 0.19%to 1.53%,extraction recoveries were(81.67±6.30)%to(90.83±3.34)%,method recoveries were(86.20±1.30)%to(114.20±1.48)%.CONCLUSION The method was confirmed to excellent specificity,sensitivity,operability and propitiousness for determination of meropenem,imipenem,concentrations in human plasma.
