摘  要:旨在分析溴结构域PHD锌指转录因子(Bromodomain PHD-finger transcription factor,BPTF)基因多态性与云上黑山羊产羔性能的关联性,以期为高繁殖力山羊新品种培育提供参考。利用Sequenom MassARRAY^■SNP技术对BPTF基因3个候选位点进行分型,探索其在云上黑山羊(YS,n=544)、济宁青山羊(JN,n=133)和辽宁绒山羊(LN,n=91)3个群体中的遗传学特征,并对其与云上黑山羊产羔性能(包括产羔数、初生窝重和断奶窝重)的关联性进行分析。结果表明,BPTF基因g.48615819 C>G位点和g.48616808 C>G位点在不同繁殖力品种(JN和LN)中的基因型分布存在极显著差异(P<0.01),而g.48616783 C>T位点在不同繁殖力品种(JN和LN)中的基因型分布无显著性差异(P>0.05)。在云上黑山羊中,BPTF基因g.48615819 C>G位点GC型个体的产羔数显著大于CC型个体(P<0.05),该位点对初生窝重和断奶窝重无显著影响(P>0.05);BPTF基因g.48616783 C>T位点和g.48616808 C>G位点对产羔数、初生窝重和断奶窝重无显著影响(P>0.05)。综上,BPTF基因g.48615819 C>G位点可作为云上黑山羊产羔数选择的潜在分子标记,但不适合窝重的选择。This study was performed to analyze the association between the polymorphism of Bromodomain PHD-finger transcription factor(BPTF)gene and kidding performance of Yunshang Black goats.The Sequenom MassARRAY^■system was used for genotyping of the three candidate loci of BPTF,of which the genetic characteristics of Yunshang Black goats(YS,n=544),Jining Grey goats(JN,n=133)and Liaoning Cashmere goats(LN,n=91)were explored,and whose association with kidding performance including litter size,litter weight at birth and at weaning was also analyzed.The genotype distributions of candidate loci(BPTF gene g.48615819 C>G,BPTF gene g.48616808 C>G)in breeds with different fecundity(JN and LN)were extremely significantly different(P<0.01).However,the genotype distributions of the candidate locus(BPTF gene g.48616783 C>T)in breeds with different fecundity(JN and LN)were not significantly different(P>0.05).The association analyses of YS showed that there was a significantly increased litter size of GC individuals compared to CC individuals for g.48615819 C>G at BPTF(P<0.05),there was no significant effects of this locus on litter weight at birth and litter weight at weaning(P>0.05),while there was no significant effects of the loci(BPTF gene g.48616783 C>T,BPTF gene g.48616808 C>G)on litter size,litter weight at birth and litter weight at weaning(P>0.05).These results suggested that the g.48615819 C>G locus at BPTF gene be used as a potential molecular marker for litter size in goats,but it was unsuitable for selection on the litter weight.
