秦皮乙素调节Lewis肺癌小鼠的免疫功能及抑瘤作用研究

摘　　要：目的研究秦皮乙素(esculetin,Esc)对Lewis肺癌小鼠免疫功能的调节作用及对CD4+CD25+Treg细胞分化的影响。方法取60只小鼠腋下接种肺癌细胞建立肺癌小鼠模型。造模成功后随机分为模型组、Esc低剂量组、Esc高剂量组和顺铂组,每组15只,另取15只作为正常组。Esc低剂量组、Esc高剂量组和顺铂组分别腹腔注射Esc 20、40 mg·kg^(-1)、顺铂5 mg·kg^(-1),正常组、模型组实验期间腹腔注射等量生理盐水。记录小鼠体质量变化;计算抑瘤率、胸腺指数和脾脏指数;用ELISA法检测脾脏组织中白细胞介素-6(interleukin-6,IL-6)、干扰素-γ(interferon-γ,IFN-γ)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的含量;用流式细胞术检测CD4+CD25+Treg细胞分化比例;用qRT-PCR检测T淋巴细胞中叉头状转录因子p3(forkhead box p3,Foxp3)和IL-10 mRNA的相对表达量。结果与模型组比较,Esc低剂量组、Esc高剂量组的体质量、胸腺指数、脾脏指数和IL-10 mRNA的相对表达量升高,瘤质量、IL-6、IFN-γ和TNF-α蛋白的表达水平、CD4+CD25+Treg细胞分化比例及Foxp3 mRNA的相对表达量降低(P<0.05),顺铂组的体质量、瘤质量、脾脏指数、CD4+CD25+Treg细胞分化比例及IL-6、IFN-γ和TNF-α蛋白的表达水平、Foxp3 mRNA的相对表达量降低,白细胞介素-10(IL-10)mRNA的相对表达量升高(P<0.05);与Esc低剂量组比较,Esc高剂量组小鼠的瘤质量、IL-6、IFN-γ、TNF-α蛋白的表达水平、CD4+CD25+Treg细胞分化比例及Foxp3 mRNA的相对表达量降低,体质量、抑瘤率、胸腺指数、脾脏指数和IL-10 mRNA的相对表达量升高(P<0.05),顺铂组的体质量、瘤质量、胸腺指数、脾脏指数、IL-6、IFN-γ、TNF-α蛋白的表达水平、CD4+CD25+Treg细胞分化比例和Foxp3 mRNA的相对表达量降低,抑瘤率和IL-10 mRNA的相对表达量升高(P<0.05);与Esc高剂量组比较,顺铂组小鼠体质量、瘤质量、胸腺指数、脾脏指数、IL-6、IFN-γ、TNF-α蛋白Objective To study the regulatory effect of esculetin(Esc)on the immune function of in mice with Lewis lung cancer and its effect on the differentiation of CD4+CD25+Treg cells.Methods 60 mice were axillarily inoculated with lung cancer cells to establish lung cancer mice model.After successful modeling,the mice were randomly divided into model group,low dose of Esc group,high dose of Esc group and cisplatin group,with 15 mice in each group.Another 15 mice were taken as normal group.20,40 mg·kg^(-1) Esc and 5 mg·kg^(-1) cisplatin were injected intraperitoneally in low dose of Esc group,high dose of Esc group and cisplatin group respectively.The normal group and model group were injected with the same amount of normal saline during the experiment.The changes in body weight of mice were recorded.The tumor inhibition rate,thymus index and spleen index were calculated.The levels of interleukin-6(IL-6),interferon-γ(IFN-γ)and tumor necrosis factor-α(TNF-α)in spleen were detected by ELISA.The proportion of CD4+CD25+Treg cells was detected by flow cytometry.The relative expression levels of forkhead box p3(Foxp3)and IL-10 mRNA in T lymphocytes were detected by qRT-PCR.Results Compared with model group,the body weight,thymus index,spleen index and the relative expression of IL-10 mRNA were increased,and the tumor weight,the expression of IL-6,IFN-γ and TNF-α protein,the differentiation ratio of CD4+CD25+Treg cells and the relative expression of Foxp3 mRNA were decreased in low dose of Esc group and high dose of Esc group(P<0.05).The body weight,tumor weight,spleen index,the differentiation ratio of CD4+CD25+Treg cells and the expression of IL-6,IFN-γ and TNF-α protein,the relative expression of Foxp3 mRNA were decreased,and the relative expression of IL-10 mRNA was increased in cisplatin group(P<0.05).Compared with the low dose of Esc group,the tumor weight,IL-6,IFN-γ and TNF-α protein levels,CD4+CD25+Treg cell differentiation ratio and the relative expression of Foxp3 mRNA were decreased,while the body weight,
