摘 要:目的观察异鼠李素(isorhamnetin,Iso)对子宫腺肌病(adenomyosis,AM)小鼠子宫异位内膜侵袭性与子宫内膜细胞凋亡的作用,并探讨可能的机制。方法40只AM小鼠随机分为AM组,Iso低、中和高剂量组,每组10只,10只健康小鼠作为对照组。Iso低、中和高剂量组用Iso混悬液(质量浓度5 g·L^(-1)羧甲基纤维素钠)1 mL灌胃,剂量分别为25、50和100 mg·kg^(-1),对照组、AM组用等体积5 g·L^(-1)的羧甲基纤维素钠溶液灌胃。每天1次,连续干预2个月。检测血清干扰素γ(interferon-γ,IFN-γ)、白细胞介素-4(interleukin-4,IL-4)水平;用Tunel染色法检测子宫内膜细胞凋亡率;用HE染色法观察子宫异位内膜侵袭性;用免疫印迹法检测子宫内膜组织中P53-273H、上皮生长因子受体(epithelial growth factor receptor,EGFR)、p-EGFR和转录信号转导子与转录激活子3(signal transducer and activator of transcription 3,STAT3)和p-STAT3蛋白的表达量。结果与AM组比较,Iso低剂量组血清IFN-γ水平、子宫内膜细胞凋亡率升高,IL-4水平、异位内膜侵袭性评分降低(P<0.05);与Iso低剂量组比较,Iso中剂量组血清IFN-γ水平、子宫内膜细胞凋亡率升高,IL-4水平、异位内膜侵袭性评分降低(P<0.05);与Iso中剂量组比较,Iso高剂量组血清IFN-γ水平、子宫内膜细胞凋亡率升高,IL-4水平、异位内膜侵袭性评分降低(P<0.05)。与AM组比较,Iso低剂量组P53-273H蛋白表达量及p-EGFR/EGFR、p-STAT3/STAT3降低(P<0.05);与Iso低剂量组比较,Iso中剂量组P53-273H蛋白表达量及p-EGFR/EGFR、p-STAT3/STAT3降低(P<0.05);与Iso中剂量组比较,Iso高剂量组P53-273H蛋白的表达量及p-EGFR/EGFR、p-STAT3/STAT3降低(P<0.05)。结论Iso可降低AM小鼠子宫异位内膜侵袭性,促进子宫内膜细胞凋亡,推测该作用与抑制p53信号通路有关。Objective To observe the effects of isorhamnetin(Iso)on the invasiveness of ectopic endometrium and endometrial cell apoptosis in adenomyosis(AM)mice,and to explore the possible mechanism.Methods 40 AM mice were randomly divided into AM group,Iso low,medium and high dose group,with 10 mice in each group and 10 healthy mice as the control group.Iso low,medium and high dose groups were intragastricaly given Iso suspension(concentration 5 g·L^(-1) sodium carboxymethyl cellulose)1 mL at 25,50 and 100 mg·kg^(-1),respectively.The control group and AM group were gavaged with equal volume of 5 g·L^(-1) sodium carboxymethyl cellulose solution.The intervention was performed once a day for 2 months.The serum levels of interferon-γ(IFN-γ)and interleukin-4(IL-4)were detected.The apoptosis rate of endometrial cells was detected by Tunel staining.The endometrium invasions were observed by HE staining.The expression of P53-273H and epithelial growth factor receptor in endometrial tissue(EGFR),p-EGFR,signal transducer and activator of transcription 3(STAT3)and p-STAT3 protein expression was detected by western blot.Results Compared with AM group,the serum IFN-γ level and endometrial apoptosis rate in Iso low-dose group were increased,while the IL-4 level and ectopic endometrial invasion score were decreased(P<0.05).Compared with medium-dose Iso group,the serum IFN-γ level and endometrial apoptosis rate in Iso high-dose group were increased,while the IL-4 level and ectopic endometrial invasion score were decreased(P<0.05).Compared with medium-dose Iso group,the serum IFN-γ level and endometrial apoptosis rate in Iso high-dose group were increased,while the IL-4 level and ectopic endometrial invasion score were decreased(P<0.05).Compared with AM group,p-EGFR/EGFR and p-STAT3/STAT3 were decreased in Iso low-dose group(P<0.05).Compared with low-dose Iso group,p-EGFR/EGFR and p-STAT3/STAT3 protein expression levels of P53-273H protein in medium-dose Iso group were decreased(P<0.05).Compared with Iso medium-dose group,the expre
