摘  要:目的探讨小檗碱对侵袭性肺曲霉病大鼠肺部损伤的保护及对Th2细胞因子水平的影响。方法SD雄性大鼠建立侵袭性肺曲霉病大鼠模型,随机分为模型对照组、小檗碱低剂量组和小檗碱高剂量组、阳性对照组,另设健康组。经一系列干预后,检测各组大鼠肺功能变化;用HE染色观察大鼠肺功能病理学;称定胸腺和脾脏质量,计算胸腺指数及脾脏指数;用酶联免疫吸附法(ELISA)检测大鼠肺泡灌洗液中干扰素-γ(interferon-gamma,IFN-γ)、白细胞介素-4(interleukin 4,IL-4)的含量;用RT-PCR和Western blot检测大鼠肺组织T细胞转录因子(T-box expressed in T-cells,T-bet)、GATA结合蛋白3(GATA binding protein 3,GATA-3)mRNA与蛋白的表达水平。结果模型对照组、小檗碱低剂量组、小檗碱高剂量组、阳性对照组、健康组大鼠肺容积变换和静息通气量逐渐增大,肺泡灌洗液中IFN-γ的含量逐渐增多、IL-4含量逐渐减少,腺指数与脾脏指数逐渐升高(P<0.05)。模型对照组烟曲霉菌感染弥漫全肺,菌丝分布密集,向四周扩散,血管出血严重,可见大量炎性细胞浸润;与模型对照组比较,小檗碱低剂量组、小檗碱高剂量组、阳性对照组的感染灶局限于肺部边缘,菌丝散在分布,感染部分有炎性细胞浸润,出血量较少。模型对照组、小檗碱低剂量组、小檗碱高剂量组、健康组大鼠肺部T-bet mRNA与蛋白表达水平逐渐升高,GATA-3 mRNA与蛋白的表达水平逐渐下降(P<0.05)。结论小檗碱对侵袭性肺曲霉病大鼠肺部损伤有保护作用,可恢复Th1/Th2细胞因子平衡,可能通过调节T-bet/GATA3信号通路实现。Objective To explore the protection effect of berberine on lung injury in rats with invasive pulmonary aspergillosis and the effect on Th2 cytokine levels.Methods A rat model of invasive pulmonary aspergillosis was established in SD male rats,and they were randomly divided into a model control group,a low-dose berberine group,a high-dose berberine group,a positive control group,and a healthy group.A healthy group were established separately.After a series of interventions,the changes in lung function of rats in each group were detected.The pathology of lung function in rats was observed by HE staining.The mass of thymus and spleen was weighed,and the thymus index and spleen index were calculated.The interferon-γ(IFN-γ),interleukin 4(IL-4)contents in rat alveolar lavage fluid were detected by ELISA.The rat lung tissue T cell transcription factor(T-bet),GATA binding protein 3(GATA-3)mRNA and protein expression level were detected by RT-PCR and Western blot.Results In the model control group,the low-dose berberine group,the high-dose berberine group,the positive control group,and the healthy group,the lung volume was changed,the resting ventilation volume gradually increased,and the content of IFN-γin the bronchoalveolar lavage fluid gradually increased.The IL-4 content gradually decreased,and the gland index and spleen index increased gradually(P<0.05).In the model control group,the infection of Aspergillus fumigatus diffused throughout the whole lung,the mycelium was densely distributed and spread to the surrounding area,the blood vessels were hemorrhaged seriously,and a large number of inflammatory cells were infiltrated;Compared with the model control group,the infection focus of the berberine group,the high-dose berberine group,and the low-dose positive control group was limited to the edge of the lung,the hyphae were scattered,the infected part had inflammatory cell infiltration,and the amount of bleeding was less.In the model control group,the low-dose berberine group,the high-dose berberine group,and the
