摘 要:目的 观察赤芍总苷对慢性阻塞性肺疾病大鼠炎症和凋亡的影响,以及探究其分子机制。方法将40只雄性SD大鼠随机分为健康对照组、模型组、赤芍总苷高剂量组和赤芍总苷低剂量组,每组10只。采用烟熏联合气管内滴入脂多糖构建慢阻肺大鼠模型。赤芍总苷高剂量组和赤芍总苷低剂量组烟熏前1h分别给予150mg/kg和75mg/kg赤芍总苷灌胃,健康对照组和模型组给予等量的生理盐水,1次/d连续4周。HE染色观察各组大鼠肺组织病理学形态;ELISA检测大鼠血清中炎性因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素6(IL-6);TUNEL计数检测大鼠Ⅱ型肺泡细胞的凋亡;Western Blot检测大鼠肺组织中核细胞表面Toll样受体4(TLR4)、内核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、细胞凋亡相关因子B淋巴细胞瘤-2(Bcl-2)、水溶性相关蛋白(Bax)和半胱氨酸蛋白酶-3(Caspase-3)蛋白。结果 与健康对照组比较,模型组大鼠的气道狭窄,肺泡囊状扩展,TNF-α、IL-1β、IL-6的表达量显著增加(P<0.05),TLR4、NF-κB、Bcl-2、Bax和Caspase-3蛋白表达量和凋亡指数显著增加(P<0.05)。与模型组比较,赤芍总苷高剂量组和赤芍总苷低剂量组的局部肺泡稍微扩张,可见炎性细胞浸润,TNF-α、IL-1β、IL-6的表达量显著降低(P<0.05),TLR4、NF-κB、Bcl-2、Bax和Caspase-3蛋白表达量和凋亡指数显著降低(P<0.05)。结论 赤芍总苷能改善慢阻肺模型大鼠肺组织形态,抑制炎性和凋亡反应这与抑制NF-κB信号通路有关。Objective To observe the effect of total paeony glycoside(TPG) on inflammation and apoptosis in rats with chronic obstructive pulmonary disease(COPD),and explore its molecular mechanism.Methods Forty male SD rats were randomly divided into healthy control group, model group,high-dose total peony glycosides group and low-dose total peony glycosides group,with 10 rats in each group.A rat model of chronic obstructive pulmonary disease was constructed by smoking combined with instill-ation of lipopolysaccharide into the trachea.High-dose group and low-dose group were given 150 mg/kg and 75 mg/kg total peony glycosides by gavage 1 hour before smoking respectively.Healthy control group and model group were given the same amount of normal saline,once a day,for 4 consecutive weeks.HE staining was used to observe the pathological morphology of the lung tissues of rats in each group;ELISA was used to detect the inflammatory factors tumor necrosis factor-α(TNF-α),interleukin(IL)-1β,and IL-6 in rat serum;the TUNEL count detection apoptosis of rat type Ⅱalveolar cells;Western Blot was used to detect proteins of TLR4,NF-κB,TNF-α,IL-1β,IL6,Bcl-2,Bax and Caspase-3 in rat lung tissue.Results Compared with healthy control group,the airway of model group was narrowed,the alveolar sac-like expansion,the expression of TNF-α,IL-1β,and IL-6 were increased significantly(P<0.05),the protein expression and apoptosis index of TLR4,NF-κB,Bcl-2,Bax and Capsase-3 were increased significantly(P<0.05);compared with model group,the local alveoli of high-dose total peony glycosides group and low-dose total peony glycosides group were slightly dilated,indicating that inflammatory cell infiltration was seen,and the expression of TNF-α,IL-1β,and IL-6 was significantly reduced(P<0.05),the protein expression and apoptosis index of TLR4,NF-κB,Bcl-2,Bax and Capsase-3 were significantly reduced(P<0.05).Conclusion Total paeony glycosides could improve the lung tis sue morphology of COPD model rats and inhibit inflammatory and apoptotic react
