摘　　要：白喉毒素(diphtheria toxin,DT)是由感染β噬菌体基因组的白喉棒状杆菌(Corynebacterium diphtheriae)所产生的外毒素,具有极强的细胞毒性。而交叉反应物质197(cross-reacting material 197,CRM197)是DT无毒突变体。CRM197通过氨基酸突变导致其丧失细胞毒性,目前在感染性疾病的预防用疫苗开发、非感染性疾病的治疗用疫苗开发、癌症治疗等领域得到了广泛尝试和运用。现就CRM197的分子基础、异源表达以及应用现状作一概述。Diphtheria toxin(DT)is an exotoxin produced by Corynebacterium diphtheriae C7(beta)and has extremely strong cytotoxicity.Cross-reacting material 197(CRM197),a DT non-toxic mutant that contains a single amino acid substitution,has been widely adopted and applied in the development of prophylactic vaccines for infectious diseases,therapeutic vaccines for non-infectious diseases,and cancer treatment.In this paper,the molecular basis,heterologous expression and application of CRM197 are reviewed.
