摘  要:为挖掘地衣芽孢杆菌LCDD6的应用价值,将LCDD6基因组测序并进行初步分析;通过鉴定培养基检测菌株的促生功能;将LCDD6发酵液灌根处理核桃苗,60 d后测定核桃苗长势、核桃叶片的光合作用和植株系统抗性;利用Illumina Miseq高通量测序分析根际土壤的微生物菌群结构.预测到与促生、生防相关的功能基因/基因簇10个;LCDD6具有产吲哚乙酸(IAA)、铁载体、降解蛋白和解磷的能力;与LB对照组相比,处理组核桃苗株高、茎粗、分枝数分别增加了24.68%、7.03%和28.35%,植株鲜重和干重分别提高了12.96%和21.55%;叶片叶绿素含量提高了47.50%,达到极显著性差异(P<0.01),在暗适应下光系统Ⅱ的最大光化学效率提高了9.43%,达到显著性差异(P<0.05);叶片SOD、POD和CAT酶活分别增强了20.58%、26.10%和16.67%;根际土壤微生物菌群的丰富度提高,有益菌的相对含量增加.本研究表明LCDD6对核桃幼苗具有稳定的促生生防作用和丰富的基因资源,在绿色农业发展中具有重要的研究和应用价值.(图7表4参43)To determine the value of Bacillus licheniformis LCDD6 in agricultural applications,we analyzed the genome sequence of strain LCDD6.In addition,the growth-promoting function of strain LCDD6 was investigated using identification media.Walnut seedlings were treated with the fermentation broth of strain LCDD6 and grown for 60 days.The growth condition,photosynthetic capacity,and systemic resistance of walnut seedlings were determined.The microbial community structure of the rhizosphere was analyzed by Illumina MiSeq sequencing.A total of 10 genes or gene clusters were predicted according to growth promotion and biocontrol capacities.Strain LCDD6 could produce indole-3-acetic acid(IAA),siderophores,and proteases and solubilize phosphates.Compared with the control,treatment with strain LCDD6 increased the plant height,stem diameter,branch number,fresh weight,and dry weight of walnut seedlings by 24.68%,7.03%,28.35%,12.96%,and 21.55%,respectively.Under dark adaptation,the maximal efficiency of the PSII photochemistry of walnut seedlings treated with strain LCDD6 was increased by 9.43%(P<0.05).The superoxide dismutase,peroxidase,and catalase activities of walnut seedlings treated with strain LCDD6 were increased by 20.58%,26.10%,and 16.67%,respectively.The abundance of the microbial community in the rhizosphere of walnut seedlings treated with strain LCDD6 was increased.Furthermore,the relative content of beneficial bacteria and fungi was increased.Our results showed strain LCDD6 contained useful genes and had a stable growth-promoting and biocontrol effect on walnut seedlings,which could have important research and application value in sustainable agriculture.
