摘  要:类甜蛋白(thaumatin-like proteins,TLPs)是重要的病程相关蛋白,在植物抵御生物和非生物逆境中扮演着重要角色.为揭示香蕉类甜蛋白基因家族序列及表达特性,利用生物信息学方法对香蕉TLPs(MaTLPs)进行全基因组鉴定,分析MaTLPs的理化性质、亚细胞定位、系统进化和染色体定位情况以及该基因家族的基因结构和基因复制事件,并基于转录组数据研究它们在低温、高温和香蕉枯萎病菌(FocTR4)胁迫下的表达情况.结果显示:香蕉共包含40个MaTLP成员,编码长度在106-333 aa之间、相对分子量介于11779.98-34292.20、等电点为4.28-9.10的蛋白质,大部分MaTLPs定位于胞外.系统进化分析显示MaTLPs可分为Group1-9,MaTLPs在进化中发生了13次片段重复和2次串联重复事件.MEME分析结果显示,MaTLP家族成员含有10个motif,且大多数成员具有相同的motif数量和分布方式.MaTLPs启动子上含有大量逆境胁迫和植物激素相关顺式作用元件,同时还存在11种转录因子家族的结合位点.MaTLPs在低温、高温和FocTR4胁迫下呈现不同程度的响应,其中MaTLP2-2和MaTLP7-1受3种逆境处理影响显著;高温和低温处理显著抑制MaTLP5-6和MaTLP11-1的表达,显著促进MaTLP9-3的表达;MaTLP4-1的表达受低温和FocTR4影响显著,而MaTLP5-3和MaTLP6-1仅受高温显著影响.本研究表明全基因组、串联和片段重复事件三者共同作用促进MaTLP基因家族成员数量的扩展,该家族部分成员在抵抗非生物胁迫和病原菌过程中发挥作用.(图7表3参42)Thaumatin-like proteins(TLPs),a type of pathogenesis-related protein,play important roles in plant responses to both biotic and abiotic stresses.To reveal the sequence characteristics and expression patterns of the TLP gene family in bananas,a series of bioinformatics methods were used to identify the physicochemical characteristics,subcellular localization,and phylogenetic relationship of banana TLPs(MaTLPs)and to reveal the chromosomal location,gene structure,and gene replication events of MaTLP members.Moreover,their expression patterns under low-temperature,high-temperature,and Fusarium oxysporum f.sp.cubense Tropical Race 4(FocTR4)treatments were analyzed using transcriptome data.In total,40 MaTLP were identified in the banana genome.The sequence lengths of their encoded proteins were between 106 and 333 aa,their molecular weights ranged from 11779.98 to 34292.20,and their theoretical isoelectric points were between 4.28 and 9.10.Most MaTLP members were located in the extracellular space.Phylogenetic analysis showed that MaTLPs could be divided into nine groups.Gene duplication analysis identified 13 segmental duplications and two tandem duplication events in MaTLPs during banana evolution.MEME analysis showed that MaTLPs contained 10 motifs,and most MaTLPs had the same motif number and similar distribution.Promoter sequence analysis revealed that MaTLP promoters contained a large number of cis-acting elements related to light,hormone,and stress responses,as well as binding sites for 11 types of transcription factors.Gene expression analysis showed that MaTLPs responded differently to low temperature,high temperature,and FocTR4 treatments,among which MaTLP2-2 and MaTLP7-1 responded significantly to all three treatments.High-and low-temperature treatments significantly inhibited the expression of MaTLP5-6 and MaTLP11-1 but significantly induced the expression of MaTLP9-3.MaTLP4-1 was responsive to low temperatures and FocTR4 responses,whereas MaTLP5-3 and MaTLP6-1 were only significantly responsive to high te
