摘 要:目的分析Klotho蛋白及Wnt/β-catenin信号通路相关分子在子痫前期中的表达及临床意义。方法选取2021年1月至12月新疆医科大学第五附属医院收治的34例子痫前期患者作为病例组,另选取同期30例健康产妇作为对照组,分娩后采集两组胎盘组织标本,通过Western blot法检测Klotho、Wnt1、β-catenin、MMP-2、MMP-9蛋白水平,通过侵袭实验检测滋养细胞侵袭能力。结果病例组产妇的Klotho、Wnt1、β-catenin、MMP-2、MMP-9蛋白相对表达量低于对照组,差异均有统计学意义(P<0.05)。重度子痫产妇的Klotho、Wnt1、β-catenin、MMP-2、MMP-9蛋白相对表达量低于轻度子痫产妇,差异均有统计学意义(P<0.05)。病例组产妇滋养细胞侵袭能力低于对照组,重度子痫产妇滋养细胞侵袭能力低于轻度子痫产妇,差异均有统计学意义(P<0.05)。结论子痫前期产妇胎盘组织中Klotho、Wnt1、β-catenin蛋白表达水平降低,其低表达与滋养细胞侵袭能力减弱、子痫前期发生及发展存在一定关系。Objective To analyze the expression and clinical significance of Klotho protein and Wnt/β-catenin signal pathway related molecules in preeclampsia.Methods A total of 34 patients with preeclampsia treated in the Fifth Affiliated Hospital of Xinjiang Medical University from January to December 2021 were selected as the case group,and another 30 healthy pregnant women in the same period were selected as the control group.The placental tissue samples were collected from the two groups after delivery,the levels of Klotho,Wnt1,β-catenin,MMP-2 and MMP-9 protein were detected by Western blot,the invasion ability of trophoblast was detected by invasion experiment.Results The relative expressions of Klotho,Wnt1,β-catenin,MMP-2 and MMP-9 protein in the case group were lower than those in the control group,the differences were statistically significant(P<0.05).The relative expressions of Klotho,Wnt1,β-catenin,MMP-2 and MMP-9 protein in the severe eclampsia were lower than those in mild eclampsia,the differences were statistically significant(P<0.05).The invasion ability of trophoblast in the case group was lower than that in the control group,and the invasion ability of trophoblast in women with severe eclampsia was lower than that in women with mild eclampsia,the differences were statistically significant(P<0.05).Conclusion The expression level of Klotho,Wnt1 andβ-catenin protein decrease in placental tissue of preeclampsia women,low expression is related to the weakening of trophoblast invasion and the occurrence and de velopment of preeclampsia.
