摘 要:目的:通过检测粒径大小不同的黔产黄柏超微粉的粉体学性质及化学成分,为黄柏破壁饮片生产过程中粒径的选择和控制提供数据支撑。方法:将黄柏粉碎成50目、120目、200目、300目及500目等不同粒径的粉体,测定该粉体的堆密度、休止角和含水量,扫描电子显微镜(SEM)观测,再以UPLC-Q-TOF/MS技术分析该粉体的化学成分种类及含量。结果:黄柏粉体休止角均大于40度,堆密度则是粒度居中的200目粉体最大;从黔产黄柏传统饮片粉体和破壁饮片中分离鉴定出化合物18个,其中生物碱类9个、有机酸类4个、不饱和脂肪酸3个及三萜类化合物1个;生物碱类和有机酸和类化学成分的峰面值均在粉体为300目时上升至最高,随后下降;粉体过300目和500目盐酸小檗碱、盐酸黄柏碱的含量无明显差异;不饱脂肪酸类成分的峰面积值随粉体粒径变小而增加。不同目数粉体的化学成分种类无明显差异,各成分的峰面积值有较大差异。结论:结合中医临床用药和制药企业用药的实际情况进行综合考虑,建议黔产黄柏破壁饮片粉碎时粉体过300目较为适宜。下一步课题组将继续研究黄柏粉体,为黄柏破壁饮片的生产加工及储存提供数据参考。Objective:The properties and chemical components of different particle sizes of Phellodendron amurense powder were studied to provide data support for the production of broken wall decoction pieces of Phellodendron amurense and the control of particle size.Methods:The Phellodendron amurense was crushed into 50 mesh,120 mesh,200 mesh,300 mesh and 500 mesh powders by ultra-fine pulverizer.The morphology was observed by scanning electron microscope(SEM),and the bulk density,angle of repose and water content of the powders were measured.The chemical composition and content of the powders with different particle sizes were determined and analyzed by UPLC-Q-TOF/MS.Results:The repose angle of Phellodendron amurense powder is more than 40degrees,and the bulk density is the largest of 200 mesh powder with medium particle size;Eighteen compounds were isolated and identified from the common powder and broken wall decoction pieces of Phellodendron amurense,including 9 alkaloids,4 organic acids,3 unsaturated fatty acids and 1 triterpenoid;There was no significant difference in the chemical composition of powders with different mesh numbers,and the peak area values of each component were quite different.The peak values of alkaloids,organic acids and analogues increased to the highest when the powder was 300 mesh,and then decreased;The content of berberine hydrochloride and Phellodendron amurense in powder passing through 300 mesh and 500 mesh had no significant difference;The peak area value of unsaturated fatty acids increased with the increase of the number of comminuted mesh.Conclusion:Combined with the actual situation of traditional Chinese medicine clinical use and pharmaceutical enterprises,it is suggested that it is more appropriate for the powder to exceed 300 mesh when the broken wall decoction pieces of Phellodendron amurense produced in Guizhou are crushed.In the next step,the research group will continue to study the powder of Phellodendron amurense so as to provide data reference for the production,processing and
