摘 要:利用体外转录信使RNA(IVT mRNA)进行疫苗接种、基因治疗已经成为医学领域的热点。在全球SARS-CoV-2大流行期间,信使RNA(mRNA)药物研发备受关注。成熟的mRNA需要5’帽来保证mRNA的稳定和基因表达。在这项研究中,我们研究了mRNA的体外转录,探讨了不同mRNA加帽方法,包括酶法加帽、抗反向帽类似物(ARCA)加帽以及三核苷酸帽类似物Clean Cap加帽,并尝试了使用多个新型ARCA结构对m RNA进行加帽修饰,为mRNA药物制剂的研发打下了一定的基础。The use of in vitro transcribed messenger RNA(IVT mRNA) for vaccination and gene therapy has become a hot spot in the medical field.During the global SARS-CoV-2 pandemic,messenger RNA(mRNA) drug development has received much attention.Mature mRNAs require 5’ caps for gene expression and mRNA stability.In this study,we investigated mRNA transcription in vitro and explored different m RNA capping methods,including enzymatic capping,anti-reverse cap analog(ARCA) capping,and the trinucleotide cap analog Clean Cap capping.The resulted mRNA products were analyzed for the productivity,intergrity and purity.The data supports further development of mRNA based drugs for disease treatments.
