摘 要:以矾根带腋芽茎段为试验材料,研究次氯酸钠的不同处理时间对外植体出芽率和污染率的影响;以及不同激素浓度对矾根芽增殖和生根的影响;以及不同培养温度和暗培养对组培苗生长的影响。结果表明,次氯酸钠处理矾根带腋芽茎段10 min,污染率最低为27.38%,出芽率最高达94.86%,生长状态良好。MS+6-BA 0.6 mg/L+NAA 0.02 mg/L+蔗糖30 g/L是最佳的增殖培养基,增殖倍数最高达9.53倍,芽大而绿,长势较好。1/2 MS+NAA 0.2 mg/L+活性炭0.5 g/L+蔗糖30 g/L是最佳的生根培养基,生根率最高达100%,平均根数8~9条,平均根长达12~13 cm。培养温度为24~26℃,最有利于组培苗增殖和生根,增殖倍数最高达9.5倍,生根率最高达100%,苗高为3 cm左右。与光照培养对比,暗培养10 d,组培苗的生长速度显著加快,再次移到光照下培养,增殖倍数和生根率均有所提高,光暗交替培养可有效缩短生长周期,在短期内可获得大量优质组培苗。Alum root stem with axillary bud was used as experimental material to study the effects of different treatment times of sodium hypochlorite on explants germination rate and contamination rate;and the effect of different hormone concentrations on the proliferation and rooting of alum root bud;and the effects of different culture temperatures and dark culture on the growth of tissue culture seedlings.The results showed that the stem segment with axillary bud was treated with sodium hypochlorite for 10 min,the lowest pollution rate was only 27.38%,the highest budding rate was 94.86%,and the growth state was good.MS+6-BA 0.6 mg/L+NAA 0.02 mg/L+sucrose 30 g/L was the best medium for proliferation,the highest multiplication was 9.53 times,the bud was big and green,the growth was good.1/2 MS+NAA 0.2 mg/L+AC 0.5 g/L+sucrose 30 g/L was the best rooting medium,the rooting rate was up to 100%,the average number of roots was 8-9,the average length of roots was 12-13 cm.The culture temperature was 24-26℃,which was the best for the multiplication and rooting of tissue culture plantlets.The multiplication factor was 9.5 times,the rooting rate was 100%and the height of plantlets was about 3 cm.Compared with light culture,the growth rate of tissue culture seedlings was significantly accelerated after 10 days of dark culture,and the multiplication ratio and rooting rate were increased when the tissue culture seedlings were moved to light culture,a large number of high quality tissue culture plantlets could be obtained in a short time.
