摘 要:目的:探讨薏苡附子败酱散加味对盆腔炎性疾病后遗症(SPID)模型大鼠的治疗作用,初步阐述其治疗SPID可能的作用机制。方法:SPF级SD雌性大鼠60只,随机选取10只作为正常组,其余均采用混合菌悬液联合器械损伤法制备SPID大鼠模型。造模成功后将模型大鼠随机分成5组:模型组,薏苡附子败酱散加味低、中、高剂量组(以下简称低、中、高剂量组),妇科千金片组。灌胃给药21 d后,腹主动脉采血,取双侧子宫。观察大鼠血液流变学指标;测定大鼠血清中C反应蛋白(CRP)、白细胞介素(IL)-2、IL-10、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量;检测大鼠子宫组织中JNK、p38蛋白和mRNA表达水平。结果:与正常组比较,模型组大鼠各项血液流变学指标、血清中CRP、IL-2、TNF-α、MDA水平及大鼠子宫组织中JNK、p38蛋白和mRNA表达水平均显著上升(P<0.05),SOD、IL-10水平显著下降(P<0.05)。与模型组比较,高、中剂量组及妇科千金片组大鼠各项血液流变学指标显著降低(P<0.05);各给药组大鼠血清中CRP、IL-2、TNF-α水平及大鼠子宫组织中JNK、p38蛋白和mRNA表达水平显著下降(除低剂量组p38蛋白表达外)(P<0.05),IL-10、SOD水平显著上升(P<0.05)。结论:薏苡附子败酱散加味可能通过改善血液流变学指标,降低促炎症因子水平,提高抗炎症因子水平,调控机体氧化-抗氧化系统平衡,以抑制介导炎症反应的JNK/p38信号通路表达而达到治疗SPID作用。Objective:To investigate the therapeutic effect of modified Yiyi Fuzi Baijiang Powder on sequelae of pelvic inflammatory disease(SPID)model rats,and to preliminarily expound its possible mechanism of action in the treatment of SPID.Methods:Sixty SPF SD female rats were randomly selected as normal group(n=10),and the rest were treated with mixed bacterial suspension combined with device injury method to establish SPID rat model.After successful modeling,the model rats were randomly divided into 5 groups:model group,modified Yiyi Fuzi Baijiang Powder low-dose,medium-dose and high-dose groups(low-dose,medium-dose,high-dose groups),and Fuke Qianjin Tablets group.After 21 days of intragastric administration,blood was collected from the abdominal aorta,and the bilateral uterus was collected.Observation of hemorheological indexes in rats;determination of C-reactive protein(CRP),interleukin-2(IL-2),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)in serum of rats,superoxide dismutase(SOD)and malondialdehyde(MDA)contents;the protein and mRNA expression levels of JNK and p38 in rat uterine tissue were detected.Results:Compared with the normal group,various hemorheological indexes,serum CRP,IL-2,TNF-α,MDA levels and the protein and mRNA expression level of JNK,p38 in rat uterus tissue in the model group were significantly increased(P<0.05),the levels of SOD and IL-10 decreased significantly(P<0.05).Compared with the model group,various hemorheological indexes of the rats in the high and middle dose groups and the Fuke Qianjin Tablet group were significantly decreased(P<0.05),IL-2,TNF-αlevels and the protein and mRNA expression level of JNK,p38 in rat uterus tissue(except the protein expression of p38 in the low-dose group)decreased significantly(P<0.05),while IL-10 and SOD levels increased significantly(P<0.05).Conclusion:Modified Yiyi Fuzi Baijiang Powder may improve hemorheological indexes;reduce the level of pro-inflammatory factors and increase the level of anti-inflammatory factors;regulate the balance of the bo
