摘  要:目的 观察吡咯喹啉醌(PQQ)对人卵巢癌细胞SKOV-3上皮间质转化(EMT)的影响,并探讨可能机制。方法 取对数期SKOV-3细胞,随机分为Control组、PQQ组、PQQ+LiCl[Wnt/β-连环蛋白(β-catenin)通路激活剂]组。Control组不处理,PQQ组加入PQQ 16μmol·L培养24 h, PQQ+LiCl组加入PQQ 16μmol·L^(-1)培养24 h后再加入LiCl 60 mmol·L^(-1)培养24 h。以Transwell实验检测细胞侵袭能力;以划痕实验检测细胞迁移能力;以蛋白质印迹法检测细胞β-catenin、反式激活活性蛋白(c-myc)、糖原合成酶激酶3β(GSK-3β)蛋白表达量。结果 Control组、PQQ组、PQQ+LiCl组每个视野细胞数分别为(95.20±11.39),(29.40±4.25)和(46.00±5.97)个,迁移率分别为(99.58±1.24)%,(18.19±2.36)%和(62.67±7.50)%;这3组细胞β-catenin蛋白表达量分别为1.10±0.14,0.12±0.03和0.71±0.08,c-myc蛋白表达量分别为0.76±0.08,0.22±0.04和0.53±0.06,GSK-3β蛋白表达量分别为0.09±0.01,1.03±0.14和0.42±0.05。上述指标,PQQ组与Control组比较,PQQ+LiCl组与PQQ组比较,差异均有统计学意义(均P<0.05)。结论 PQQ对SKOV-3细胞的侵袭、迁移、EMT具有抑制作用,可能与抑制Wnt/β-catenin信号通路活性有关。Objective To observe the effect of pyrroloquinoline quinone(PQQ) on epithelial-mesenchymal transition(EMT) of human ovarian cancer cell SKOV-3, and to explore the possible mechanism. Methods SKOV-3 cells in log phase were randomly divided into control group, PQQ group, PQQ+LiCl(Wnt/β-catenin pathway activator) group. The control group was not treated. The PQQ group was cultured with PQQ 16 μmol·L^(-1)for 24 h, and the PQQ+LiCl group was cultured with PQQ 16 μmol·L^(-1)for 24 h, followed by LiCl 60 mmol·Lfor 24 h. The cell invasion ability were detected by Transwell test. The cell migration ability were detected by scratch test. The cell β-catenin, transactivation active protein(c-myc), glycogen synthase kinase 3β(GSK-3β) protein expression were detected by Western blotting. Results The number of cells in each field of view in the control group, PQQ group, and PQQ+LiCl group were(95.20±11.39),(29.40±4.25),(46.00±5.97), respectively, migration rate were ( 99. 58 ± 1. 24) %,( 18. 19 ± 2. 36) %,( 62. 67 ± 7. 50) %,the β-catenin protein expression of cells were1. 10 ± 0. 14,0. 12 ± 0. 03,0. 71 ± 0. 08,c-myc protein expression were 0. 76 ± 0. 08,0. 22 ± 0. 04,0. 53 ± 0. 06,GSK-3β protein expression were 0. 09 ± 0. 01,1. 03 ± 0. 14,0. 42 ± 0. 05. Compared between PQQ group and control group,PQQ + LiC l group and PQQ group,the differences of the above indicators were statistically significant( all P < 0. 05). Conclusion PQQ has inhibitory effects on the invasion,migration and EMT of SKOV-3 cells,which may be related to the inhibition of Wnt/β-catenin signaling pathway activity.
