摘 要:目的 研究酒女贞子对综合激怒法致大鼠肝肾阴虚证的肝肾保护作用。方法 雄性SD大鼠随机分为5组:正常组、模型组及低、中、高3个剂量实验组,每组8只。用综合激怒方法建立大鼠因情志失调致肝肾阴虚模型。造模后,低、中、高3个剂量实验组分别给予0.1,0.2和0.4 g·kg^(-)的酒女贞子提取物,qd,连续14 d,正常组和模型组给予蒸馏水。用酶联免疫吸附法检测大鼠血清中内分泌指标促甲状腺激素(TSH)和睾酮(T)的含量及肝肾组织中白细胞介素-6(IL-6)和白细胞介素-1β(IL-1β)水平,试剂盒微量法检测肝肾组织匀浆液中氧化应激指标丙二醛(MDA)和还原型谷胱甘肽(GSH)活性。结果 正常组、模型组和高剂量实验组大鼠的血清TSH含量分别为(14.02±0.74),(15.55±0.90)和(14.38±0.73) mU·L^(-);这3组血清的T含量分别为(262.82±14.55),(245.28±9.570)和(272.86±18.91) pg·mL^(-);这3组肝肾组织的IL-6含量分别为(106.10±2.78),(111.63±3.66)和(103.73±4.17) pg·mL^(-);这3组肝肾组织的IL-1β含量分别为(25.31±0.98),(26.79±0.95)和(24.76±1.11) pg·mL^(-);这3组肝肾组织的MDA含量分别为(9.65±2.18),(13.29±2.04)和(8.41±0.9) nmol·mg^(-);这3组肝肾组织的GSH含量分别为(3.70±0.77),(1.85±0.94)和(3.57±0.56) pg·mg^(-);模型组与正常组比较,差异均有统计学意义(均P<0.05);实验组与模型组相比,差异均有统计学意义(均P<0.05)。结论 酒女贞子可通过改善氧化应激、抑制炎症因子的释放和调节内分泌紊乱对激怒致肝肾阴虚大鼠有一定的改善调节作用。Objective To investigate the protective effect of wine steaming of Ligustri lucidi Fructus on the liver and kidney of syndrome of liver and kidney Yin deficiency induced by comprehensive irritation method. Methods Male SD rats were randomly divided into 5 groups: normal group, model group and low, medium and high dose experimental groups, with 8 rats in each group. Rat models of liver and kidney Yin deficiency induced by anger were established by comprehensive anger method. After modeling, the low, medium and high dose experimental groups were given 0.1, 0.2, and 0.4 g·kgwine steaming of Ligustrum lucidum Fructus extract, qd, 14 d, and the normal group and the model group were given distilled water. The levels of thyroid stimulating hormone( TSH) and testosterone( T) in serum;interleukin-6( IL-6) and interleukin-1β( IL-1β)in liver and kidney of rats were detected by enzyme linked immunosorbent assay. The activities of malondialdehyde( MDA) and reduced glutathione( GSH) in liver and kidney homogenate were detected by kit micro method.
