丝素蛋白对斑马鱼高血脂模型的降血脂作用探究

摘　　要：为了探究丝素蛋白对斑马鱼高血脂模型是否具有抑制作用,从而拓展丝素蛋白在人类医药领域的作用,文章通过建立斑马鱼幼苗高血脂模型,并用组织染色与免疫荧光染色对模型进行检验与分析,最后脱水染色的结果通过图像软件ImageJ和SPSS 22.0进行比较分析。结果表明:随着培养基中蛋黄粉质量分数提高,斑马鱼尾脂荧光强度逐渐提高,斑马鱼高血脂症状加剧,可以建立高血脂模型。通过综合比较分析,实验造模选择含0.1%蛋黄粉的培养基。斑马鱼幼苗的高血脂模型在丝素蛋白溶液质量浓度为1.0μg/mL时,丝素蛋白中的限制性氨基酸对斑马鱼高血脂降低最明显。最后通过与市面上的降血脂药物(辛伐他汀)在同种质量浓度下比较,验证了各个质量浓度的丝素蛋白对斑马鱼高血脂模型均有抑制作用。丝素蛋白通过降低斑马鱼血浆中脂质水平来抑制高血脂,未来可用于降血脂药物替代或者作为辅药添加。Existing lipid-lowering drugs have poor efficacy,toxic and side effects or high costs,so it is quite essential to develop new lipid-lowering drugs.Silk fibroin can promote the expression of a variety of factors,and degraded amino acids can be absorbed and degraded by host animals without side effects.Zebrafish have the characteristics of small size,high spawning capacity,rapid development,transparent embryos and larvae,and have a high degree of homology with the human genome.Thus,zebrafish can be used as a model of hyperlipidemia.In order to explore whether zebra protein has inhibitory effect on zebrafish hyperlipidemia model,the role of silk fibroin in the field of human medicine is expanded.The model was tested and analyzed by tissue staining and immunofluorescence staining,and the results of the final dehydrated staining were compared by image software ImageJ and SPSS 22.0 analysis.The results show that with the increase of egg yolk powder concentration in the culture medium,the fluorescence intensity of zebrafish tail fat gradually increases,the symptoms of hyperlipidemia in zebrafish are aggravated,and a hyperlipidemia model can be established.Through comprehensive comparative analysis,the medium containing 0.1%egg yolk powder was selected for this experimental molding.In the hyperlipidemia model of zebrafish fry,when the concentration of fibroin solution is 1.0μg/mL,the restriction amino acids in the silk fibroin are most obvious for the reduction of hyperlipidemia in zebrafish.Finally,by comparing with the lipid-lowering drug(simvastatin)on the market at the same concentration,it is verified that each concentration of silk has an inhibitory effect on the zebrafish hyperlipidemia model.Silk fibroin inhibits hyperlipidemia by lowering lipid levels in zebrafish plasma and can be used in the future as a replacement of lipid-lowering drugs or as an adjunct.The role of silk fibroin has an important role in the field of medicine,and zebrafish are a good model animal.After exploring the hyperlipidemia model,it is
