摘 要:为探究晋北地区部分养殖场腹泻犊牛中大肠杆菌耐药表型与所携带耐药基因和整合子的关系,本研究采集晋北地区3个规模场腹泻犊牛肛拭子27份进行大肠杆菌分离鉴定,采用K-B法和PCR检测分离株药物敏感性和耐药基因、整合子-基因盒的携带情况,采用SPSS软件进行相关性分析。结果显示,24株大肠杆菌分离株对6类15种常用抗菌药物呈现不同程度的耐药,对β-内酰胺类抗菌药头孢噻肟、头孢唑林和氨苄西林耐药的菌株均达95.83%(23/24),但对氧氟沙星耐药的菌株只占12.50%(3/24);多重耐药菌株占比70.83%(17/24),4重耐药菌株占比最高,为33.33%(8/24);β-内酰胺类、氨基糖苷类、磺胺类、喹诺酮类、氯霉素类和四环素类耐药基因均有检出,喹诺酮类耐药基因GyrB检出率为100%;Ⅰ类整合子检出率为79.17%,最主要的基因盒排列为dfrA17-aadA5,Ⅱ、Ⅲ类整合子未被检出;耐药基因bla_(TEM)、bla_(CTX-M-1)、sul1、sul2、tetA、tetB、cat与耐药表型符合率均为100%;整合子阳性株的多重耐药表型的发生率极显著高于整合子阴性株(P<0.01)。综上,晋北地区部分养殖场犊牛源大肠杆菌分离株多重耐药情况严重,耐药表型与所携带耐药基因和Ⅰ类整合子密切相关。This study investigated the relationship between drug resistance phenotype and drug resistance gene/integron in Escherichia coli from diarrhea calves in northern Shanxi.A total of 27 anal swabs were collected from diarrhea calves in 3 farms in northern Shanxi for E.coli isolation and identification,for detection of drug sensitivity and drug resistance genes,integrons-gene cassettes by K-B method and PCR,and for correlation analysis by SPSS software.The results showed that 24 E.coli isolates had varying degrees of drug resistance to 6 classes of 15 commonly used antibiotics,of which 95.83%(23/24)were resistant toβ-lactams cefotaxime,cefazolin and ampicillin,but only 12.50%(3/24)were resistant to ofloxacin;70.83%(17/24)were multidrug-resistant,and 33.33%(8/24)were four-drug-resistant;β-lactam,aminoglycoside,sulfonamide,quinolone,chloramphenicol and tetracycline resistance genes were all detected,with quinolone resistance gene GyrB identified in all isolates;class I integron found in 79.17%isolates,and the major gene cassette was dfrA 17-aadA 5,however,class II and III integrons were not detected;the coincidence rate of drug resistance genes bla_(TEM),bla_(CTX-M-1),sul 1,sul 2,tetA,tetB,cat and drug resistance phenotype was 100%;the incidence of multiple drug resistance phenotype of integron-positive strains was significantly higher than that of integron-negative strains(P<0.01).Thus,multiple drug resistant E.coli is present in calves from some farms in northern Shanxi,and the drug resistance phenotype is closely related to the drug resistance genes and class I integron.
