摘  要:为了优化黄芩苷铜的合成工艺并阐明其对猪源大肠杆菌K88的抗菌作用,本试验以反应时间、反应温度、五水硫酸铜/黄芩苷的摩尔比和碳酸氢钠/黄芩苷的摩尔比为4个考察因素,以黄芩苷铜转化率为考查指标,进行正交试验设计,优化得到黄芩苷铜的合成工艺为6.2 mmoL黄芩苷加入水中,在60℃滴加7.44 mmoL碳酸氢钠水溶液,搅拌均匀后加入9.3 mmoL五水硫酸铜水溶液混合均匀,反应75 min,再加入甲醇搅拌得黄芩苷铜。本试验进一步分析了黄芩苷铜对猪源大肠杆菌K88超微结构和大肠杆菌K88培养上清液中不耐热肠毒素释放量的影响,结果显示,黄芩苷铜的浓度为750μg/mL和375μg/mL时,分别极显著(P<0.01)和显著(P<0.05)抑制猪源大肠杆菌K88不耐热肠毒素的释放;当黄芩苷铜的浓度为375μg/mL时,可引起细胞质明显流失,导致细菌死亡。结果表明,黄芩苷铜有望作为一种新型中兽药应用于仔猪肠道疾病的防治。This study investigated the optimal synthesis of baicalin-copper and its antibacterial effect on porcine Escherichia coli K88.Synthesis of baicalin-copper was optimized by taking account of reaction time,reaction temperature,molar ratio of copper sulfate pentahydrate to baicalin,molar ratio of sodium bicarbonate to baicalin,and using baicalin conversion rate as reference.The optimum synthesis process of baicalin-copper was found as follows:6.2 mmoL baicalin was added into water,7.44 mmoL sodium bicarbonate aqueous solution was added dropwise at 60℃,9.3 mmol copper sulfate pentahydrate aqueous solution was added,after 75 min,added methanol and obtained baicalin copper.The results showed that the two concentrations of baicalin-copper(750μg/mL and 375μg/mL)extremely significantly(P<0.01)and significantly(P<0.05)inhibited the release of heat labile enterotoxin from porcine E.coli K88;375μg/mL baicalin-copper leading to significant loss of cytoplasm and bacterial death.In conclusion,baicalin-copper has potential to be explored as a new Chinese veterinary drug for the prevention and treatment of intestinal diseases in piglets.
