摘 要:目的:建立基于昼夜规律的阿霉素诱发大鼠肾源性水肿模型,为蒙药利尿作用机制研究提供模型基础。方法:设早、晚8点为模型制备时间,以SD大鼠为研究对象,分为早、晚6.5 mg·kg^(-1)两组,分别于造模第1、8天,大鼠尾静脉注射阿霉素6.5(4.0+2.5)mg·kg^(-1)。观察注射阿霉素后大鼠的一般状态及尿量;于造模第4周末麻醉并处死大鼠,计算肾脏指数,测定血清生化指标含量。结果:造模后,从第2天开始两组大鼠不同程度腹泻,精神状态萎靡,毛发光泽变差、脱毛;从第5天开始个别大鼠出现红色尿,食欲不振,体重增长缓慢;从第8天开始个别大鼠出现尾巴溃烂现象;与正常组比较,模型组肾脏指数均有上升;早、晚模型组大鼠造模后第1天、第14天及第28天时尿量均明显少于早、晚正常组(P<0.05);但第14天时晚模型组血清ADH、ALD、AQP1及AQP2含量显著高于早模型组(P<0.05),指标变化稳定性优于早模型组。结论:经昼夜节律造模后发现,阿霉素诱导肾病大鼠模型具有明显的昼夜规律,晚8时造模,造模周期可缩短到14天。该研究为蒙药利尿作用机制分析提供了模型基础。Objective:To establish a model of nephrogenic edema induced by Adriamycin in rats based on circadian rhythm,and to provide a model basis for study of the diuretic mechanism of Mongolian medicine.Methods:The model preparation time was set at 8 o’clock in the morning or evening,SD rats were taken as research object,and they were divided into two groups:a morning group and an evening group.On the 1st and 8th day of modeling,the rats were injected with Adriamycin 6.5(4.0+2.5)mg·kg^(-1)via tail vein.The general state and urine volume of rats were observed after Adriamycin injection.At the end of the 4th week of modeling,the rats were anesthetized and sacrificed.The renal index was calculated and the contents of serum biochemical indexes were measured.Results:From the second day after modeling,the rats of two groups had diarrhea in different degrees,their mental states were dispirited,and their hair luster became poor and their hair were depilated;From the 5th day,some rats in the two groups showed red urine,anorexia and slow weight gain;From the 8th day,some rats had tail ulceration.Compared with the control group,the renal index of each model group increased.The rats’urine volume in the morning group and the evening group was significantly lower than that in the control model group on the 1st,14th and 28th day after modeling(P<0.05).The contents of ADH,ALD,AQP1 and AQP2 in rats’serum in the evening group were significantly higher than those in the morning model group on the 14th day after modeling(P<0.05),and the stability of index changes was better than that in the morning group.Conclusion:Through circadian rhythm modeling,it is found that the rat model of Adriamycin induced nephropathy has obvious circadian law.At 8 PM modeling,the modeling period could be shortened to 14 days.This study provides a model basis for the diuretic mechanism analysis of Mongolian medicine.
