摘  要:为探讨促性腺激素抑制激素(GnIH)对雄性SD大鼠肝脏脂代谢的影响,以不同浓度的GnIH(0、1和10μg/100μL,总体积200μL,溶解于生理盐水中)早晚2次定点给SD大鼠腹腔注射。持续注射6周后,观察记录GnIH对雄性SD大鼠的表观影响,并进一步检测血液生化指标、肝脏脂代谢相关蛋白表达以及腺苷酸活化蛋白激酶(AMPK)信号通路的影响。结果表明:与对照组相比,腹腔注射不同浓度的GnIH均可导致雄性SD大鼠采食量显著增多(P<0.05),平均日增重显著加快(P<0.05),Lee指数显著增加(P<0.05),血液中甘油三酯浓度显著升高(P<0.05),而高密度脂蛋白极显著下降(P<0.01)。进一步研究发现,腹腔注射不同浓度的GnIH均可使肝脏中脂肪酸合成酶(FASN)蛋白表达显著升高(P<0.05),而激素敏感脂肪酶(HSL)蛋白表达显著降低(P<0.05),脂联素(ADPN)和过氧化物酶体增殖物激活受体γ(PPARγ)蛋白表达均极显著降低(P<0.01),CAAT区/增强子结合蛋白(C/EBP)表达极显著升高(P<0.01)。与此同时,腹腔注射不同浓度的GnIH均可使肝脏中AMPKα的磷酸化水平极显著降低(P<0.01),但AMPKβ的磷酸化水平无显著差异(P>0.05)。研究表明,腹腔注射GnIH可能通过抑制AMPKα信号通路从而抑制肝脏脂肪酸分解,同时促进肝脏脂肪合成,导致雄性大鼠肝脏脂肪的沉积。To investigate the effect of GnIH on liver lipid metabolism in male SD rats, the rats were intraperitoneally injected with different concentrations of GnIH(0, 1 and 10 μg/100 μL, total volume 200 μL, dissolved in normal saline) twice a day, once in the morning and once in the evening. After continuous injection for 6 weeks, the apparent effects of GnIH on the male SD rats were observed and recorded, and the effects of blood biochemistry, liver lipid metabolism related protein expression and AMPK signal pathways were further detected. The results showed that, compared with the control group, intraperitoneal injection of different concentrations of GnIH resulted in a significant increase in the feed intake of the rodents(P<0.05), in their average daily gain(P<0.05), in their Lee index(P<0.05), in triglyceride concentration in blood(P<0.05) and an extremely significant decrease in their high density lipoprotein(P<0.01). Further study showed that intraperitoneal injection of different concentrations of GnIH significantly increased the expression of the FASN protein(P<0.05), significantly reduced the expression of the HSL protein(P<0.05), extremely significantly reduced the expression of the ADPN and PPARγ proteins in the liver of the rats(P<0.01), and extremely significantly increased the expression of the C/EBP protein(P<0.01). At the same time, intraperitoneal injection of different concentrations of GnIH extremely significantly reduced the phosphorylation level of AMPKα in the liver(P<0.01), but there was no significant difference in the phosphorylation level of AMPKβ(P>0.05). The above results suggested that intraperitoneal injection of GnIH might inhibit hepatic fatty acid decomposition by inhibiting the AMPKα signaling pathway, promoted hepatic fat synthesis, and finally led to hepatic fat deposition in male rats.
