摘  要:目的探究姜油酮对高脂饮食诱导的非酒精性脂肪性肝病(not-alcoholic fatty liver disease,NAFLD)大鼠的保护作用及其机制。方法采用高脂饲料饲养构建NAFLD大鼠模型,SD大鼠分为对照组、模型组、姜油酮Ⅰ组和姜油酮Ⅱ组,姜油酮Ⅰ组和姜油酮Ⅱ组每日分别给予0.12 mg·kg^(-1)和0.24 mg·kg^(-1)姜油酮1次,对照组和模型组给予等量碳酸氢钠溶液。酶联免疫吸附法检测甘油三酯(TG)、总胆固醇(TC)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)水平;HE染色观察肝脏组织病理变化;蛋白质免疫印记法检测肝脏组织中Toll样受体4(TLR4)和核因子E2相关因子2(Nrf2)蛋白水平;实时荧光定量PCR法检测肝脏组织中TLR4 mRNA和Nrf2 mRNA水平。结果与模型组比较,姜油酮可以改善NAFLD大鼠肝脏损伤,降低ALT、AST及MDA水平(P<0.05),升高GSH、SOD水平(P<0.05),肝组织中TLR4及TLR4 mRNA较模型组降低(P<0.05),Nrf2及Nrf2 mRNA较模型组升高(P<0.05)。结论姜油酮对NAFLD大鼠有一定保护作用,其机制可能与调节TLR4和Nrf2有关。Objective To investigate the protective effect and mechanism of zingerone on high fat diet induced non-alcoholic fatty liver disease(NAFLD)in rats.Methods A rat model of NAFLD was established by high-fat diet.SD rats were divided into control group,model group,zingerone I group and zingerone II group.Zingerone I group and zingerone II group were given 0.12 mg·kg^(-1)and 0.24 mg·kg^(-1)zingerone daily once,respectively.The control group and the model group were given the same amount of sodium bicarbonate solution.The levels of triglyceride(TG),total cholesterol(TC),alanine aminotransferase(ALT),aspartate aminotransferase(AST),superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione(GSH)were measured by ELISA.HE staining was used to observe the pathological changes of liver tissue;Toll-like receptor 4(TLR4)and nuclear factor E2 related factor 2(Nrf2)protein levels in liver tissue were detected by Western-Blot;TLR4 and Nrf2 gene levels in liver tissue were detected by real-time fluorescence quantitative PCR.Results Compared with the model group,zingerone could improve liver injury,decrease ALT,AST and MDA levels(P<0.05),increase GSH and SOD levels(P<0.05),and TLR4 and TLR4 mRNA in liver tissue were lower than the model group(P<0.05),and Nrf2 and Nrf2 mRNA were higher than the model group(P<0.05).Conclusion Zingerone has a protective effect on NAFLD rats,and its mechanism may be related to the regulation of TLR4 and Nrf2.
