摘  要:目的探讨舒芬太尼(sufentanil,SUF)对胶质瘤细胞恶性行为的影响及其可能作用机制。方法体外培养人胶质瘤细胞U251,实验分组:Con组、SUF-L组、SUF-M组、SUF-H组、阳性对照组;CCK-8法、划痕实验、Transwell实验与平板克隆形成实验分别检测细胞增殖、迁移、侵袭及克隆形成;采用qRT-PCR法检测胶质瘤组织、癌旁组织与各组U251细胞中miR-3614-5p的表达量;miR-NC、miR-3614-5p mimics分别转染至U251细胞,anti-miR-NC、anti-miR-3614-5p分别转染至U251细胞后加入100μg·L^(-1)舒芬太尼处理细胞24 h,采用上述方法分别检测细胞增殖、克隆形成、迁移及侵袭。结果与Con组比较,SUF-L组、SUF-M组、SUF-H组细胞增殖抑制率和miR-3614-5p的表达量升高,细胞克隆形成数和侵袭细胞数减少,划痕愈合率降低,且呈剂量依赖性;SUF-L组、SUF-M组、SUF-H组与阳性对照组各指标比较差异均有统计学意义(P<0.05);与癌旁组织比较,胶质瘤组织中miR-3614-5p的表达量降低;转染miR-3614-5p mimics后细胞增殖抑制率升高,细胞克隆形成数和侵袭细胞数减少,划痕愈合率降低,而转染anti-miR-3614-5p可降低SUF对U251细胞生物学行为的作用。结论舒芬太尼可通过促进miR-3614-5p表达而减弱胶质瘤细胞增殖、克隆形成、迁移及侵袭能力。Objective To explore the effect of sufentanil(sufentanil,SUF)on the malignant behavior of glioma cells and its possible mechanism.Methods Human glioma cells U251 were cultured in vitro,and the experimental groups were divided into Con group,SUF-L group,SUF-M group,SUF-H group and positive control group.CCK-8 method,scratch test,Transwell test and plate clone formation test were applied to estimate cell proliferation,migration,invasion and clone formation.qRT-PCR method was performed to assess the expression of miR-3614-5 p in glioma tissues,adjacent tissues and U251 cells in each group.miR-NC and miR-3614-5 p mimics were transfected into U251 cells respectively.Anti-miR-NC and anti-miR-3614-5 p were respectively transfected into U251 cells and treated with 100μg·L^(-1)sufentanil for 24 h.The above methods were used to detect cell proliferation,clone formation,migration and invasion.Results Compared with the Con group,the cell proliferation inhibition rate and the expression of miR-3614-5 p in the SUF-L,SUF-M,and SUF-H groups were increased,while the number of cell clones formed cells and the number of invasive cells were decreased,and the rate of scratch healing was decreased,and was in a dose-dependent manner.There were significant differences in each index between the SUF-L group,SUF-M group,SUF-H group and the positive control group(P<0.05).miR-3614-5 p expression in glioma tissues was reduced compared with adjacent tissues.After transfection of miR-3614-5 p mimics,the cell proliferation inhibition rate was increased,the clone formation number and invasive cell number were decreased,and the scratch healing rate was decreased.However,transfection with anti-miR-3614-5 p could reduce the effect of SUF on the biological behavior of U251 cells.Conclusion Sufentanil could reduce the proliferation,clone formation,migration and invasion of glioma cells by promoting the expression of miR-3614-5 p.
