摘 要:为解析AglaRpS8功能特征及在光肩星天牛生长发育过程中的作用机制,通过克隆核糖体蛋白基因AgRpS8的ORF全长序列,采用生物学信息方法分析AglaRpS8基因的理化性质、结构及功能特征;利用荧光定量PCR检测该基因在不同发育阶段的表达特征。结果表明:AglaRpS8基因ORF区长度627 pb,编码个208氨基酸,无跨膜区螺旋结构,具6个Motif结构域,该基因等电点5.21,分子量大小为49.99 KDa,与AmRpS8基因同源性最高,与TcRpS8-like和TcRpS8基因亲缘性最近。AglaRpS8基因在光肩星天牛不同发育阶段存在显著的差异性表达,在幼虫1~2龄期,3~4龄期,5龄期相对表达量较平稳,在光肩星天牛成虫期相对表达量较低,而在光肩星天牛雌虫交配后怀卵的阶段相对表达量达到最高值。In order to analyze the functional characteristics of AglaRpS8 and its mechanism of action in the growth and development of A.glabripennis,we cloned the full-length ORF sequence of the ribosomal protein gene AgRpS8,and analyzed the physicochemical properties,structure and functional characteristics of AglaRpS8 using biological information methods.Fluorescence quantitative PCR detected the spatiotemporal expression characteristics of AglaRpS8.The results showed that the ORF region of AglaRpS8 was 627 pb in length,encoded 208 amino acids,had no helical structure in the transmembrane region,and had 6 Motif domains.The gene had an isoelectric point of 5.21 and a molecular weight of 49.99 KDa,which had the highest homology with AmRpS8,with the closest affinity with TcRpS8-like and TcRpS8.AglaRpS8 is expressed to different degrees in different developmental stages of A.glabripennis.The relative expression level of AglaRpS8 is stable in the 1st-2nd instar,3rd-4th instar,and 5th instar larvae,and the relative expression level is lower in the adult stage.The relative expression level reached the highest value in the developmental stage of gestation after mating.The results revealed that AglaRpS8 plays an important role in the physiological process of egg formation after mating in A.glabripennis females,laying a foundation for the analysis of the potential physiological and biological functions of AglaRpS8.
