摘  要:目的探讨人参皂苷Rb1(GRb1)激活羟甲基戊二酰辅酶A还原酶降解蛋白1(HRD1)通路对动脉粥样硬化介导的内皮细胞凋亡的影响。方法将人主动脉内皮细胞(HAECs)分为正常对照组(NC组)、模型组[氧化低密度脂蛋白(ox-LDL),100 mg/L]、GRb1组(50μmol/L)、GRb1+si-NC组(si-HRD1阴性对照)和GRb1+si-HRD1组,比较各组HAECs的细胞活力、活性氧(ROS)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平、细胞凋亡、B细胞淋巴瘤-2(Bcl-2)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)、B细胞淋巴瘤-2相关X蛋白(Bax)及HRD1蛋白表达。结果与NC组比较,模型组细胞活力、SOD水平、Bcl-2及HRD1蛋白降低,ROS、MDA水平、细胞凋亡率、Caspase-3及Bax蛋白升高,差异有统计学意义(P<0.05);与模型组比较,GRb1组细胞活力、SOD水平、Bcl-2及HRD1显著升高,ROS、MDA水平、细胞凋亡率、Caspase-3及Bax蛋白降低,差异有统计学意义(P<0.05);沉默HRD1逆转了GRb1对ox-LDL诱导的动脉粥样硬化内皮细胞的影响。结论GRb1可能通过激活HRD1通路抑制ox-LDL诱导的动脉粥样硬化内皮细胞凋亡。Objective To investigate the effects of ginsenoside Rb1(GRb1)activated hydroxymethylglutaryl-CoA reductase degradation protein 1(HRD1)signaling pathway on atherosclerosis-mediated endothelial apoptosis.Methods Human aortic endothelial cells(HAECs)were divided into normal control group(NC group),model group(oxidized low-density lipoprotein,100 mg/L),GRb1 group(50μmol/L),GRb1+si-NC group(si-HRD1 negative control)and GRb1+si-HRD1 group.Following items were compared:cellular viability,reactive oxygen species(ROS),superoxide dismutase(SOD),malondialdehyde(MDA)levels,apoptosis,B-cell lymphoma-2(Bcl-2),cysteamine Caspase-3(Caspase-3),B-cell lymphoma-2 related X protein(Bax),and HRD1 protein expression.Results Compared with the NC group,the cellular viability,SOD level,Bcl-2 and HRD1 protein in the model group significantly reduced;levels of ROS and MDA,apoptosis rate,Caspase-3,and Bax protein significantly increased,differences were statistically significant(P<0.05);compared with model group,cellular viability,SOD level,Bcl-2,and HRD1 protein in GRb1 group significantly increased;ROS,MDA level,apoptosis rate,Caspase-3,and Bax protein significantly decreased,differences were statistically significant(P<0.05);silencing HRD1 reversed the effect of GRb1 on ox-LDL-induced atherosclerotic endothelial cells.Conclusion GRb1 might activate the HRD1 pathway to inhibit apoptosis of atherosclerotic endothelial cells induced by ox-LDL.
