摘 要:目的探讨牛蒡苷元(Arc-G)对小鼠止血、血小板数目、凝血、体内血栓形成及血块回缩功能的影响。方法将小鼠随机分为空白对照组(生理盐水)、阿司匹林组(100 mg/kg)、Arc-G低剂量组(12.5 mg/kg)和Arc-G高剂量组(50 mg/kg),剪断小鼠尾尖、观察各组小鼠断尾出血时间;下腔静脉取血,检测血小板数目、血小板平均体积(MPV)、凝血酶原时间(PT)、部分凝血活酶时间(APTT)、凝血酶时间(TT)和纤维蛋白原(FIB);腹腔注射角叉菜胶建立鼠尾部血栓模型,计量鼠黑尾比例,观察Arc-G对血栓形成的影响;通过血块回缩实验,观察Arc-G体外抑制血栓形成的作用。结果阿司匹林组、Arc-G低剂量组和Arc-G高剂量组小鼠断尾出血时间较空白对照组显著延长,差异有统计学意义(P<0.01或P<0.05);与空白对照组比较,Arc-G低剂量组和Arc-G高剂量组的血小板数目、MPV、PT、APTT、TT及FIB比较,差异均无统计学意义(P>0.05);鼠尾部血栓模型结果显示,阿司匹林组、Arc-G高剂量组黑尾比例明显低于空白对照组(P<0.01),差异有统计学意义;与溶剂组比较,Arc-G(3.125μmol/L)和Arc-G(12.5μmol/L)可抑制血块回缩,差异有统计学意义(P<0.05)。结论Arc-G具有抑制止血和抗血栓的作用。Objective To investigate the effects of Arctigenin(Arc-G)on hemostasis,platelet count,clotting,thrombosis and clot retraction in mice.Methods Mice were randomly divided into control group(saline),aspirin group(100 mg/kg),Arc-G low dose group(12.5 mg/kg),Arc-G high dose group(50 mg/kg).The tail tip of the mouse was cut off,and the tail bleeding time of all groups was observed.Blood was taken from the inferior vena cava,and platelet count,mean platelet volume(MPV),prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT),and fibrinogen(FIB)were tested.The tail thrombosis model was established by intraperitoneal injection of carrageenan to count black tail percentage study the effect of Arc-G on thrombosis;the role of Arc-G in inhibiting thrombus formation was observed by clot retraction experiment.Results The bleeding time of mice in the aspirin group,the Arc-G low-dose group and the Arc-G high-dose group was significantly longer than that of blank control group,differences were statistically significant(P<0.01 or P<0.05).Platelets,MPV,PT,APTT,TT,and FIB in the low-dose Arc-g group and the high-dose Arc-g group were not significantly different from the blank control group(P>0.05).Tail thrombosis model indicated that the percentage of black tail in aspirin group,the Arc-g high-dose group was significantly lower than that of blank control group,differences were statistically significant(P<0.01).Compared with the solvent group,Arc-G(3.125μmol/L),and Arc-G(12.5μmol/L)could significantly inhibit clot retraction,differences were statistically significant(P<0.05).Conclusion Arc-G has the function of inhibiting hemostasis and thrombosis.
