摘 要:通过线粒体调控区D-loop基因部分序列对广东沿岸和三沙海域3个地理群体(广东惠东海龟国家级自然保护区、广东粤西和海南省三沙市)共132个绿海龟样本进行遗传多样性研究。结果表明,3个绿海龟地理群体的D-loop基因片段序列的A+T含量为68.2%,碱基组成具有明显的偏向性;共定义了19个单倍型,单倍型多样性在0.157~0.775之间,核苷酸多样性在0.00063~0.01017之间;3个群体间存在一定的分化,其中三沙群体和其他2个群体存在高度的分化,粤西群体和海龟保护区群体存在较小的分化,3个群体之间存在基因交流现象;采用邻位连接法(NJ)对132个序列片段和19个单倍型分别构建了系统发育树,3个群体的绿海龟存在较为复杂的交叉聚类现象,19个单倍型聚类为3个大的分支。本研究结果表明本研究的绿海龟总体样本具有较高的遗传多样性,广东沿岸与三沙海域的绿海龟种群存在基因交流现象。本研究为探究我国绿海龟种群遗传多样性和开展绿海龟种质资源保护奠定理论基础。The genetic diversity of Chelonia mydas in 3 geographical populations were studied by partial sequences of Mitochondrial D-loop gene from 132 samples which collected from Huidong Sea Turtle National Reserve,the western Guangdong Province and the Sansha sea area.The results showed that A+T contents of D-loop gene fragments was 68.2%and the base composition showed obvious bias in 3 geographical populations.A total of 19 haplotypes were detected in 132 samples,and their haplotype diversity was between 0.157-0.775,and their nucleotide diversity was between 0.00063-0.01017.There were high levels of genetic differentiation between Sansha populations and the other two populations,and low levels of genetic differentiation between the western Guangdong Province populations and National Huidong Sea Turtle Nature Reserve populations.The gene flow was existed between 3 geographical populations.According to the neighbour-joining tree of 132 D-loop partial sequences and 19 haplotypes,there were complex cross-clustering among 3 populations,and it were respectively divided 19 haplotypes into 3 clades.The result suggested that the 132 green sea turtles samples had a high genetic diversity.
