摘 要:目的分析益生菌大肠埃希菌Nissle 1917(EcN)鞭毛蛋白(FliC)的结构,并预测其B和T细胞表位。方法从GenBank数据库中获取EcN FliC的氨基酸序列。应用ProtParam在线工具分析EcN FliC理化特性,Protscale在线工具分析亲疏水性,PSIPRED软件及DNAstar软件预测分析二级结构,I-TASSER在线软件预测三级结构,DNAstar在线工具中Protean软件和ABCpred软件综合预测B细胞抗原优势表位,细胞毒性T细胞(cytotoxic T cell,CTL)表位预测网站预测T细胞表位。结果EcN FliC的相对分子质量为60937.57,化学分子式为C_(2584)H_(4201)N_(745O940)S_(6),不稳定指数为15.23;亲水性平均值为-0.324,为亲水性蛋白;α螺旋及无规则卷曲是EcN FliC二级结构中的主要结构;预测的三级结构模型具有正确的结构拓扑,可信度高;共筛选出13个B细胞优势表位及3个CTL优势表位。结论EcN FliC特征分析及表位预测结果为其作为免疫佐剂的应用奠定了基础。Objective To analyze the structure of probiotic Escherichia coli Nissle1917(EcN)flagellin(FliC)and predict its B and T cell epitopes.Methods The amino acid sequence of EcN FliC was obtained from the GenBank database and analyzed for physicochemical property by ProtParam online tool,for hydrophilicity and hydrophobicity by Protscale online tool,for secondary structure by PSIPRED and DNAstar software,and for tertiary structure by I-TASSER online software.Meanwhile,the prominent epitope of B cell antigen was predicated by the Protean software in DNAstar online tool and ABCpred tool,while the T cell epitope by cytotoxic T cell(CTL)epitope prediction website.Results EcN FliC was a hydrophilic protein with a hydrophilic value of-0.324 and a relative molecular mass of 60937.57,of which the chemical molecular formula was C_(2584)H_(4201)N_(745O940)S_(6),and the instability coefficient was 15.23.The secondary structure of EcN FliC was mainly composed ofα-helix structure and random coil,while the predicted tertiary structure showed correct structural topology with high reliability.A total of 13 dominant B cell epitopes and 3 dominant CTL epitopes were screened.Conclusion The characteristic analysis and epitope prediction of EcN FliC laid a foundation of application of EcN FliC as an immune adjuvant.
