摘  要:目的研究蒸黄精对肾阴虚大鼠肠道菌群的影响,并结合代谢组学筛选出粪便差异代谢物。方法将SD雄性大鼠适应性喂养5 d后随机分为7组,连续灌胃给药21 d后收集大鼠粪便,用于16S rDNA测序分析及代谢组学分析。结果肾阴虚大鼠肠道内菌群增加主要表现在厚壁菌门下的毛螺菌科Allobaculum菌和毛螺菌-NK4A136-group的富集,OPLS-DA计量结果表明,黄精提取物可以改变肾阴虚大鼠粪便代谢轮廓。与模型对照组相比,炮制组共发现丙酸、马来酸、苹果酸、甘氨酸、L-亮氨酸、尿嘧啶、L-缬氨酸、L-异亮氨酸等11个差异代谢物存在显著性差异;所涉及到的代谢途径有氨酰tRNA生物合成、丙酮酸代谢、丙氨酸、天冬氨酸和谷氨酸代谢、抗坏血酸和醛酸代谢等。结论蒸黄精可以改善肾阴虚大鼠的肠道菌群,主要通过调节氨基酸类和有机酸类物质改善肾阴虚症状,毛螺菌科丰度的降低可能是黄精治疗肾阴虚的作用机制。Objective To study the effect of steamed Polygonatum on the intestinal microflora of rats with kidney-yin deficiency,and to screen differential metabolites in feces using metabolomics.Methods Male SD rats were randomly divided into seven groups after five days of adaptive feeding.Feces were collected after 21 days of continuous intragastric administration for 16S rDNA sequencing and metabolomics analysis.Results The increase of intestinal flora in rats with kidney-yin deficiency was mainly manifested in the enrichment of the Allobaculum and Spirobacterium-NK4A136-group of the Family Officidae under the Phylum pachychocetes.The results of OPLS-DA showed that Polygonatum extract could change the fecal and stool metabolic profile of rats with kidney-yin deficiency.Compared with the model control group,11 metabolic differences were found in processing group,including propionic acid,maleic acid,malic acid,glycine,L-leucine,uracil,L-valine and L-isoleucine.The metabolic pathways involved include aminoacyl tRNA biosynthesis,pyruvate metabolism,alanine,aspartic acid and glutamate metabolism,ascorbic acid,aldehyde acid metabolism and other metabolic pathways.Conclusion Steaming Polygonatum can improve the intestinal microflora of rats with kidney Yin deficiency,and improve the symptoms of kidney Yin deficiency mainly by regulating amino acids and organic acids.The decrease of the abundance of Lachnospiraceae may be the mechanism of Polygonatum in treating kidney Yin deficiency.
