摘 要:目的:应用网络药理学方法探究益气化瘀补肾方治疗骨延迟愈合的潜在药效作用机制。方法:在TCMSP数据库中获取益气化瘀补肾方的主要化学成分,通过String数据库获取药物作用靶点,并在Genecards数据库、OMIM数据库及TTD数据库中获取骨延迟愈合的疾病靶点,后使用Cytoscape 3.7.2软件构建药物-靶点网络,使用拓扑学分析此网络获得核心活性成分,使用String数据库绘制靶点蛋白交互作用网络,使用拓扑学分析此网络获得核心靶点,采取Bioconductor数据库对潜在作用靶点进行GO功能及KEGG通路富集分析,探讨潜在作用靶点可能具有的生物学功能及通路。结果:从益气化瘀补肾方中筛选出251个活性成分,核心成分56个;治疗延迟愈合的靶点84个,核心靶点24个;GO分析结果显示生物过程1980个、细胞组分60个和分子功能128个;KEGG通路富集分析结果显示信号通路133条。结论:本研究初步证实益气化瘀补肾方治疗骨延迟愈合具有多成分、多靶点的特点,为进一步阐释其药理作用机制奠定了理论基础。Objective:To explore the potential pharmacological mechanism of Yiqihuayubushen recipe in the treatment of delayed bone healing by using the method of network pharmacology.Methods:The main chemical components of Yiqihuayubushen recipe were obtained from TCMSP database,the drug action targets were obtained from String database,and the disease targets of delayed bone healing were obtained from Genecards database,OMIM database and TTD database.The drug-target network was constructed by Cytoscape 3.7.2 software,the core active components were obtained by topological analysis of the network,and the target protein interaction network was drawn by String database.The core targets were obtained by topological analysis of the network,and the potential targets were analyzed by GO function and KEGG pathway enrichment analysis based on Bioconductor database to explore the possible biological functions and pathways of potential targets.Results:We screened 251 active ingredients,56 core ingredients,84 targets for the treatment of delayed healing,and 24 core targets from Yiqihuayubushen recipe,GO analysis result showed 1980 biological processes,60 cell components and 128 molecular functions,and KEGG pathway enrichment analysis showed 133 signal pathways.Conclusion:This study preliminarily confirmed that Yiqihuayubushen recipe has the characteristics of multi-components and multi-targets in the treatment of delayed bone healing,which lays a theoretical foundation for further explaining its pharmacological mechanism.
