摘 要:对疑似鸽新城疫(Newcastle disease,ND)的病料进行病毒鉴定和分离培养,获得1株鸽新城疫病毒(Newcastle disease virus,NDV)Ⅵ-HZ株,为进一步了解病毒生物学特性及其分子特征,对其进行致病性和遗传进化分析。结果表明,该毒株MDT为80 h,ICPI为1.2,EID_(50)为10^(7.0)/0.1 mL,HA效价为8 log_(2),属于中等毒力型NDV;人工感染鸽第7天出现明显腹泻和神经症状,死亡率为60%,病死鸽剖检可见脑膜充血、水肿及肠道出血;病理切片结果显示病死鸽脑组织水肿、小肠绒毛上皮细胞脱落坏死、黏膜固有层有炎症。全基因组测序显示Ⅵ-HZ株全长为15192 nt,基因排列方式为3'-NP-P/V/W-M-F-HN-L-5',其中F蛋白裂解位点氨基酸序列为^(112)KRQKRF^(117),具有NDV强毒的分子特征;系统发育进化分析表明该Ⅵ-HZ株为ClassⅡ类基因Ⅵ型毒株,与Ⅵ.2.2.2毒株亲缘关系最近。本研究为近期鸽NDV的分子流行病学调查提供了科学依据。A pigeon Newcastle disease virus(NDV)Ⅵ-HZ strain was isolated and identified from the sample of suspected Newcastle disease(ND).To further understand the biological characteristics and molecular characteristics ofⅥ-HZ strain,virus pathogenicity and genetic evolution were analyzed.The results showed that the values of MDT,ICPI,EID_(50) and Hemagglutinin(HA)are 80 h,1.2,10^(7.0)/0.1 mL and 8 log_(2),belonging to mesogenic NDV.The artificially infected pigeons showed obvious diarrhea and neurological symptoms at 7 days post-infection(dpi),and the mortality rate was 60%.The experimental pigeon autopsy showed meningeal congestion,edema and intestinal bleeding.The histology results showed brain edema,necrosis of the villous epithelial cell loss and inflammation of lamina propria.The complete genome of Ⅵ-HZ strain is 15192 nucleotides(nts)with the gene order of 3'-NP-P/V/W-M-F-HN-L-5'.The Ⅵ-HZ strain contains the ^(112)KRQKRF^(117) cleavage site in F protein,a feature generally associated with virulent NDV strains.The phylogenetic tree of full F gene sequence showed that Ⅵ-HZ strain was closely related to Class Ⅱ genotype Ⅵ NDV,and closest relationship with genotype Ⅵ.2.2.2strain.This study provides a basis for the recent molecular epidemiological investigation of pigeon NDV.
