摘  要:利用昆虫细胞-杆状病毒表达系统构建了含有H3N8亚型马流感病毒HA、NA和M1蛋白的病毒样颗粒(virus-like particles,VLPs)。VLPs的鉴定结果表明,3种蛋白正确组装出了类似于天然病毒粒子的形态结构,且具有血凝活性。将VLPs免疫马匹制备高免血清,分别设计小鼠先被动免疫再攻毒以及先攻毒再紧急注射免疫血清,结果证明马高免血清对H3N8亚型马流感病毒感染具有良好的预防和治疗效果,为研制H3N8亚型马流感VLP疫苗提供了理论依据。In order to develop an H3 N8 subtype equine influenza vaccine,the virus-like particles(VLPs)comprising of HA,NA and M1 proteins of H3 N8 equine influenza virus(EIV)were prepared by using the insect cell-baculovirus expression system.The identification results of VLPs showed that these three proteins correctly assembled the morphological structure similar to that of a natural virus particle and the assembled structure had hemagglutination activity.The hyperimmune serum from horses immunized with this VLPs were injected into mice by means of artificial passive immunization and then challenged,or challenged following by injecting hyperimmune serum.The results confirmed that the equine hyperimmune serum had good preventive and therapeutic effects on H3 N8 EIV infection,which provided data support for the development of H3 N8 equine influenza VLP vaccine.
