摘  要:基于形态学和分子生物学方法对1头秦岭羚牛肺线虫样本进行了种属鉴定。首先利用蠕虫完全剖检法分离该秦岭羚牛肺脏及支气管的肺线虫,初步观察记录虫体基本形态,进一步利用乳酚透明液对虫体进行处理后利用显微镜观察这些线虫的特征性结构,最后对虫体的ITS-2 rRNA基因进行扩增、克隆、测序以及序列分析。结果发现,从该头秦岭羚牛肺脏及支气管获得214份虫体样本,采集的新鲜虫体样本均呈淡黄色,虫体的头部、食道和尾部结构与原圆科线虫典型结构特征一致;获得了总长度为409~431 bp的ITS-2 rRNA基因,序列分析发现本研究所得虫体序列的相似性为97.5%~100.0%,系统进化分析发现本研究所得虫体单独位于一支,且与原圆科变圆属线虫(KJ439597)位于姊妹支,但与原圆科变圆属线虫的序列差异为24.1%~25.4%。结果表明,该头秦岭羚牛肺部线虫可能为原圆科变圆属线虫的一个新种,命名为羚牛变圆线虫。研究结果为秦岭羚牛肺线虫病的流行病学调查和防控提供了基础数据。Lungworms,a kind of parasitic nematodes in lungs and bronchia of human and animals,mainly cause respiratory distress,cough and other symptoms of respiratory system,leading to interstitial pneumonia,bronchitis,and seriously threatening health and life of human and animals.In March 2019,a large number of parasitic nematodes were found in lungs of a dead Budorcas taxicolor bedfordi(B.taxicolor bedfordi)in Zhouzhi County,Shaanxi Province.In order to clarify the taxa status of these nematodes,species identification of these lungworms was conducted by using morphology and molecular biological methods.Firstly,the lungworms were isolated by using complete necropsy from lungs and bronchus of B.taxicolor bedfordi,and the basic morphology of these nematodes was observed and recorded.Then,the characteristic structures of these nematodes were observed by using a microscopy after treatment with the lactophenol transparent solution.Finally,the ITS-2 rRNA gene was amplified,cloned,sequenced and analyzed.The results showed that 214 samples were collected from the lungs and bronchus of B.taxicolor bedfordi,and all worms were pale yellow in color.The cephalic extremity,esophagus and caudal extremity of the lungworms were consistent with the typical structure of nematodes within Protostrongylidae.Fragments of 409-431 bp were obtained for the ITS-2 rRNA gene.Sequence analysis showed that the sequence similarities among these nematodes were 97.5%-100.0%.Phylogenetic analysis showed that all worms isolated were clustered in a solid clade,and belonged to Varestrongylus sagittatus.However,the sequence differences between lungworms in the present study and V.sagittatus were 24.1%-25.4%(KJ439597).These results indicated that the lungworms isolated from B.taxicolor bedfordi might be a new species of Varestrongylus,named as Varestrongylus budortaxicolor sp.nov.This study provides basic data for epidemiological investigation and control of pulmonary nematodiasis of B.taxicolor bedfordi.
