摘 要:本研究为探究淫羊藿苷(ICA)对顺铂(CDDP)诱导的小鼠急性肾损伤(AKI)的保护作用及其潜在作用机制,将40只C57BL/6小鼠随机分对照组(Control),ICA组,CDDP组,淫羊藿苷治疗组(CDDP+CIA),每组10只。Control组腹腔注射等剂量0.9%生理盐水,CIA组连续灌胃20 mg/kg CIA 7 d,CDDP组一次性腹腔注射13 mg/kg CDDP,CDDP+CIA组在一次性腹腔注射CDDP 5 d后连续灌胃20 mg/kg CIA 7 d。每两天记录小鼠体重,标准试剂盒检测小鼠血清中肌酐(Scr)及尿素氮(BUN)水平,苏木精&伊红(H&E)染色检测小鼠肾脏病理变化,酶联免疫吸附(ELISA)法检测小鼠肾脏中炎性因子水平,蛋白质印迹(Western blot)法检测肾脏中核因子(NF-B)信号通路相关蛋白水平。结果显示:相比于Control组,CDDP组小鼠体重及肾脏指数显著降低,死亡率、血清中Scr和BUN水平及肾脏炎性因子水平显著上升;H&E染色结果表明小鼠肾小管出现炎性病变,管腔内出现炎性细胞浸润,红细胞增加,且CDDP组中小鼠肾脏中NF-B信号通路相关蛋白表达增加,而CIA治疗可以显著降低由CDDP诱导的小鼠肾损伤,降低NF-B信号通路相关蛋白表达,差异均有统计学意义(P<0.05)。淫羊藿苷可通过降低由NF-B信号通路介导的炎症反应来缓解顺铂诱导的小鼠急性肾损伤。Absrtact: Investigation of the protective effects of icariin(ICA) on cisplatin(CDDP)-induced acute kidney injury(AKI) in mice, 40 C57 BL/6 mice were divided into four groups: control group randomly, ICA group, CDDP group and CDDP+CIA group(n=10). The control group was treatment with an equal dosage of 0.9% normal saline intraperitoneally, the CIA group was gavage administered 20 mg/kg CIA for 7 days, and the CDDP group was intraperitoneally treatment with 13 mg/kg CDDP. In the CDDP+CIA group, 20 mg/kg CIA was administered orally for 7 days after intraperitoneal injection of CDDP for 5 days. The body weight of the mice was recorded every two days. The levels of serum creatinine(Scr) and urea nitrogen(BUN) of mice were detected by standard kits. The pathological changes of mice kidney were detected by Hematoxylin & Eosin(H&E) staining. The inflammatory cytokines levels of mice kidney were detected by enzyme-linked immunosorbent assay(ELISA). The protein expression related to NF-B signaling pathway was detected by Western blotting. The results showed that compared with the control group, the body weight and kidney index of the mice in the CDDP group were decreased significantly.The mortality, the levels of Scr and BUN, and the levels of mice kidney inflammatory factors were increased significantly. In addition, inflammatory lesions were appeared in the renal tubules of mice. Moreover, the expression of NF-B signaling pathway-related proteins in the kidneys of mice in the CDDP group was increased. However, CIA treatment could cut down CDDP-induced renal injury in mice and inhibit the NF-B signaling pathway(P<0.05) significantly. Icariin alleviates cisplatin-induced acute kidney injury in mice via reducing the inflammatory response mediated by the NF-B signaling pathway.
