摘　　要：目的:建立黄芪-丹参药对中紫草酸、丹酚酸B、丹参素、迷迭香酸、芒柄花黄素、丹参酮Ⅱ_(A)的含量测定方法。方法:采用Diamonsil Plus C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱测定黄芪-丹参药对中紫草酸、丹酚酸B、丹参素、迷迭香酸的含量,检测波长280 nm,柱温30℃,进样量10μl,流速1 ml·min^(-1)。以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱测定黄芪-丹参药对中芒柄花黄素、丹参酮Ⅱ_(A)的含量,检测波长254 nm,柱温为室温,进样量10μl,流速1ml·min^(-1)。结果:紫草酸、丹酚酸B、丹参素、迷迭香酸、芒柄花黄素、丹参酮Ⅱ_(A)在49.4~247μg·ml^(-1),706~3530μg·ml^(-1),50.2~251μg·ml^(-1),50.4~252μg·ml^(-1),0.8~4μg·ml^(-1),24~120μg·ml^(-1)范围内线性关系良好,r≥0.999。结论:此方法测操作简单、数据准确、重复性好,为进一步研究黄芪-丹参药对对多种疾病的治疗作用奠定了基础。Objective:To establish an HPLC method for the determination of purple acid,salvianolic acid B,danshensu,rosmarinic acid,formononetin and tanshinoneⅡ_(a) in drug pair Radix Astragali-Salvia miltiorrhiza.Methods:Using a Diamonsil Plus C_(18) column(250 mm×4.6 mm,5μm),and acetonitrile-0.1%formic acid aqueous solution as the mobile phase,gradient elution was used to determine the contents of purple acid,salvianolic acid B,danshensu and rosmarinic acid in Radix Astragali-Salvia miltiorrhiza.The detection wavelength was 280 nm,the column temperature was 30℃,the injection volume was 10μl,and the flow rate was 1 ml·min^(-1).Using acetonitrile-0.1%formic acid aqueous solution as the mobile phase,the contents of anthocyanin and tanshinoneⅡ_(a) in Radix Astragali-Salvia miltiorrhiza were determined by gradient elution.The detection wavelength was 254 nm,the column temperature was room temperature,the injection volume was 10μl,and the flow rate was 1 ml·min^(-1).Results:Shikoric acid,salvianolic acid B,danshensu,rosmarinic acid,formononetin and tanshinoneⅡ_(a) were linear within the range of 49.4-247μg·ml^(-1),706-3530μg·ml^(-1),50.4-252μg·ml^(-1),0.8-4μg·ml^(-1) and 24-120μg ml^(-1),respectively(r≥0.999).Conclusion:This method has the advantages of simple operation,accurate data and good repeatability,which lays foundation for further study on the therapeutic effects of Radix Astragali-Salvia miltiorrhiza on many diseases.
