摘 要:目的研究积雪草苷对砷暴露所致小鼠脂肪肝肝损伤的保护作用。方法选择60只SD小鼠,随机分为积雪草苷组、模型组、对照组,各20只,模型组、积雪草苷组均建立砷暴露所致脂肪肝肝损伤模型,对照组常规饮水,持续8周。而后给予积雪草苷组小鼠积雪草苷处理,并给予模型组、对照组小鼠等量生理盐水处理,持续4周。对比三组小鼠肝功能、肝指数,肝组织诱导型一氧化氮合成酶(iNOS)及环氧合酶-2(Cox-2)蛋白表达,肝组织白细胞介素-1β(IL-1β)、IL-6、肿瘤坏死因子-α(TNF-α)表达水平,以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)表达差异。结果积雪草苷组、模型组小鼠均成功建立砷暴露所致脂肪肝肝损伤模型,积雪草苷组脂肪肝严重程度低于模型组(P<0.05)。积雪草苷组TC、TG、ALT、AST及肝指数,肝组织iNOS、Cox-2蛋白表达,肝组织IL-1β、IL-6、TNF-α水平均高于对照组,模型组TC、TG、ALT、AST及肝指数,肝组织iNOS、Cox-2蛋白表达,肝组织IL-1β、IL-6、TNF-α水平均高于积雪草苷组(P<0.05)。积雪草苷组肝组织SOD、CAT、GPx水平均低于对照组,模型组肝组织SOD、CAT、GPx水平均低于积雪草苷组(P<0.05)。结论积雪草苷能够通过抑制炎症反应、减轻氧化应激反应,缓解砷暴露所致小鼠脂肪肝肝损伤。Objective To study the protective effect of asiaticoside on fatty liver injury induced by arsenic exposure in mice.Methods 60 SD mice were randomly divided into asiaticoside group,model group and control group,with 20 mice in each group.The model group and asiaticoside group established the model of fatty liver injury caused by arsenic exposure,while the control group drank water regularly for 8 weeks.Then mice in the asiaticoside group were treated with asiaticoside,and mice in the model group and control group were treated with the same amount of normal saline for 4 weeks.The liver function,liver index,the expression of inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(Cox-2)protein in liver tissue,the expression levels of interleukin-1β(IL-1β),IL-6,tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD)and catalase in liver tissue were compared among the three groups of mice.Results Both asiaticoside group and model group mice successfully established the model of fatty liver injury caused by arsenic exposure,and the severity of fatty liver in asiaticoside group was lower than that in model group(P<0.05).TC,TG,ALT,AST and liver index in asiaticoside group,the expression of iNOS and Cox-2 protein in liver tissue,and the levels of IL-1β,IL-6 and TNF-αin liver tissue were higher than those in control group.TC,TG,ALT,AST and liver index,the expression of iNOS and Cox-2 protein in liver tissue,and the expression of IL-1βand IL-6 in liver tissue in model group were higher than those in control group.The levels of SOD,CAT and GPx in the liver tissue of asiaticoside group were lower than those in the control group,and those in the model group were lower than those in the asiaticoside group(P<0.05).Conclusion Centella asiatica glycoside can alleviate the fatty liver injury caused by arsenic exposure in mice by inhibiting inflammation and alleviating oxidative stress.
