摘 要:应用SEA恒温反应体系对落选短体线虫进行快速检测。通过改进热裂解法提取落选短体线虫DNA,可快速完成线虫DNA释放,满足SEA恒温反应体系要求。在反应开始前加入扩增显色染料,实现扩增结果肉眼可视化判定。实验结果表明,基于SEA技术的落选短体线虫检测方法耗时短、灵敏度高、特异性强、反应条件简单,为该线虫的鉴定及口岸检疫提供了新思路。The SEA isothermal reaction system is used for the rapid detection of Pratylenchus neglectus. Extraction of Pratylenchus neglectus DNA by improved thermal splitting method can release of nematode DNA quickly and meet the requirements of SEA isothermal reaction system. Before the reaction, the amplification chromogenic dye is added to realize the visual judgment of amplification results. The experimental results show that the detection method of Pratylenchus neglectus based on SEA technology has the advantages of less time consumption, high sensitivity, specificity and simple reaction conditions, which provides a new idea for the identification and rapid detection of the nematode at ports.
