涂布镍钛合金表面的人血管内皮细胞生长因子重组质粒转染初步研究

摘  要:目的制备含重组质粒涂层,提高介入医疗器材生物相容性和耐腐蚀性,降低再狭窄率。方法将人脐静脉内皮细胞分别接种到涂布基质胶+黏连蛋白和基质胶+黏连蛋白+人血管内皮细胞生长因子(VEGF)重组质粒的镍钛合金丝上,37℃培养48 h后用正置荧光显微镜观测。结果基质胶+黏连蛋白包被的镍钛合金丝表面细胞无绿色荧光,含人VEGF重组质粒包被的镍钛合金丝表面细胞有绿色荧光,表明人VEGF重组质粒转染成功。结论人VEGF重组质粒转染成功,此种涂层具有可行性,可用于介入医疗器材表面改性。Objective To prepare a coating containing recombinant plasmid,to improve the biocompatibility and corrosion resistance of interventional medical devices,and to reduce the restenosis rate.Methods Human umbilical vein endothelial cells were separately inoculated onto nitinol alloy wires coated with matrigel+fibronectin or coated with matrigel+fibronectin+recombinant plasmid of human vascular endothelial cell growth factor(VEGF),which were cultured in an incubator at 37℃ for 48 hours,and then were examined under positive fluorescence microscope.Results No green fluorescence was observed on the surface cells of nitinol alloy wire coated with Matrigel,but on the surface cells of nitinol alloy wire coated with recombinant plasmid containing human VEGF green fluorescence was observed,indicating that human VEGF recombinant plasmid was successfully transfected.Conclusion The recombinant plasmid of human VEGF has been successfully transfected.The preparation of this kind of coating is feasible,and it can be used for the surface modification of interventional medical appliances.
