摘  要:目的:设计提取甘草多糖的工艺,并筛选最优的工艺参数。方法:以纯化水为溶剂,采用超声辅助提取工艺,再用Sevag法去除蛋白得到甘草多糖;以苯酚-硫酸法测定提取物中的多糖含量,计算甘草多糖的提取率,并以此为指标,对工艺过程中涉及选用不同粒径的甘草粉末、不同溶剂、液固比、浸泡温度、浸泡时间及超声时间做了研究,确定影响参数和参数范围。结果:甘草多糖的提取工艺,最佳参数为:原料为过筛孔为0.425 mm筛网的甘草粉末、溶剂为1%氯化钠溶液、液固比30∶1~40∶1(mL/g)、浸泡温度80℃、浸泡时间5 h、超声时间20~25 min,多糖提取率可达到4.7%以上。结论:本甘草多糖提取工艺较传统的乙醇提取工艺具有安全,环保,低成本的优点,并且多糖提取率也其相当,可广泛适用于甘草多糖的提取。Objective:To design the extraction process of glycyrrhiza polysaccharide and optimization for the process parameters.Methods:The Solvent of this process was using purified water.The protein of polysaccharide was removed by sevag method.The content of polysaccharide was determined by phenol-sulfuric acid method and the extraction rates were obtained by calculation.Taking this as an index,some parameters were studied,such as the different particle sizes of licorice powder,the different solvents,liquid-solid ratio,soaking temperature,soaking time and ultrasonic time.Finally,the optimal process parameters were selected.Results:The optimum extraction parameters of glycyrrhiza polysaccharide were as follows:the licorice powder of passing 60 mesh screen as raw material,1%sodium chloride solution as solvent,liquid-solid ratio 30∶1~40∶1,soaking temperature 80℃,soaking time 5 h and ultrasonic time 20~25 min.The extraction rate of glycyrrhiza polysaccharide was more than 4.7%.Conclusion:Comparing with the traditional ethanol extraction process,this process not olso has the advantages of safety,environmental protection and low cost,but also the polysaccharide extraction rate is equivalent.It can be widely used for the extraction of glycyrrhiza polysaccharide.
