灰飞虱G蛋白偶联受体基因的鉴定与分析

摘  要:[目的]利用灰飞虱(Laodelphax striatellus)基因组和转录组数据鉴定灰飞虱G蛋白偶联受体(G protein-coupled receptor,GPCR)基因。[方法]构建灰飞虱基因组和转录组本地数据库,以黑腹果蝇(Drosophila melanogaster)和豌豆蚜(Acyrthosiphon pisum)的GPCR基因为参照序列进行本地BLAST,去除冗余和不含多跨膜螺旋结构的序列后获得灰飞虱GPCR基因;利用NCBI CD-Search对灰飞虱GPCR序列进行跨膜结构域分析;利用Clustal W进行序列比对,并用MEGA 6软件构建灰飞虱GPCR系统发育树。[结果]鉴定了114个灰飞虱GPCR基因,它们分属于A、B、C和D这4个家族。灰飞虱A家族的GPCR数量最多,尤其是其中的神经肽和蛋白激素受体亚家族的基因数量达55个。与黑腹果蝇的GPCR相比,灰飞虱GPCR不仅种类丰富,而且有基因重复和可能的缺失现象。[结论]灰飞虱具有较为全面的GPCR系统,本研究完善了灰飞虱GPCR基因的信息,为进一步研究灰飞虱GPCR的功能奠定了基础。[Objectives]G protein-coupled receptor(GPCR)genes of Laodelphax striatellus were identified using published L.striatellus genome and transcriptome data.[Methods]A local database of the genome and transcriptome of L.striatellus was constructed.The GPCR genes of Drosophila melanogaster and Acyrthosiphon pisum were used as reference sequences for local BLAST.The GPCR genes of L.striatellus were obtained after removing the redundant sequences and the sequences without multi-transmembrane helix structure.The transmembrane domains of L.striatellus GPCR were analyzed by NCBI CD-Search.The GPCR sequences were compared using Clustal W,and the phylogenetic trees were constructed by MEGA 6 software.[Results]In this paper,114 GPCR genes of L.striatellus were identified,which belonged to A,B,C and D families.Among them,the number of family A was the largest,especially the number of neuropeptide and protein hormone receptor subfamily genes was up to 55.Compared with the GPCR of D.melanogaster,the GPCR of L.striatellus were not only rich in species,but also had gene duplications and possible deletions.[Conclusions]L.striatellus had a comprehensive GPCR system.This work perfected the information of L.striatellus GPCR genes and laid a foundation for further study of the function of L.striatellus GPCR.
