摘 要:目的 探讨长托宁联合米非司酮对子宫肌瘤大鼠子宫损伤的改善作用及可能的机制。方法50只SD大鼠随机分成对照组、模型组、长托宁组、米非司酮组及长托宁联合米非司酮组,每组10只。HE染色观察子宫组织形态学变化;ELISA方法检测血清炎症因子、雌激素和孕激素水平;TUNEL方法检测子宫组织细胞凋亡;Western blot检测Wnt信号通路相关蛋白的表达。结果 与对照组相比,模型组大鼠子宫组织形态学病理变化明显,血清TNF-α水平升高,IL-6和IL-2水平降低,血清雌二醇、孕激素和黄体生成素水平升高;子宫组织细胞凋亡水平降低;Wnt5b和β-catenin蛋白表达水平升高。长托宁联合米非司酮治疗改善子宫肌瘤大鼠子宫组织形态,降低血清TNF-α、雌二醇、孕激素和黄体生成素水平,升高IL-6、IL-2、子宫组织细胞凋亡水平,降低Wnt5b和β-catenin蛋白表达水平。结论 长托宁联合米非司酮减轻子宫肌瘤大鼠子宫损伤,其机制可能与抑制Wnt/β-catenin信号通路有关。Objective To explore the ameliorative effect of penehyclidine hydrochloride combined with mifepristone on uterine injury in rats with uterine fibroids and its possible mechanism. Methods Fifty SD rats were randomly divided into control group, model group, penehyclidine hydrochloride group, mifepristone group and penehyclidine hydrochloride combined with mifepristone group, with 10 rats in each group. HE staining was used to observe the morphological changes of uterine tissue. Levels of inflammatory cytokines in serum, estrogen and progesterone were detected by ELISA. Apoptosis of cell in uterine tissue was detected by TUNEL assay. The expression of Wnt signaling pathway related proteins was detected by Western blot. Results Compared with the control group, the uterine histological and pathological changes of model group rats were obvious, the levels of TNF-α in serum was increased, the levels of IL-6 and IL-2 was decreased, and the levels of estradiol, progesterone and luteinizing hormone in serum was increased. The apoptotic of cells in uterine tissue was decreased. The expression of Wnt5b and β-catenin was increased. Penehyclidine hydrochloride combined with mifepristone improved the morphology of uterine tissue, decreased the levels of TNF-α, estradiol,progesterone and luteinizing hormone in serum, increased the levels of IL-6, IL-2, cells apoptosis of uterine tissue,decreased and expression of Wnt5b and β-catenin in uterine fibroids rats. Conclusion Penehyclidine hydrochloride combined with mifepristone alleviates uterine injury in rats with uterine fibroids, and the mechanism of action may be related to inhibite Wnt/β-catenin signaling pathway.
